| Objective: To analyze the characteristic of the Chinese medicine that is used to prevent from and cure amnesia and summarize the basic principles for making syndrome differentiation and curing amnesia in Chinese medicine. To offer more ways and reliable literature material preventing from and curing MCI for Chinese medicine. To detect the method of estimating the curative effect in curing MCI with Chinese medicine by observing clinical curative effect and security of Shenwu Capsule in curing MCI .Methods: In the second part, we read literatures, extracted what we needed, and then input in computer. We established electronic table in Access software, and carried out literatures-extracting and counting with senior filter in Access. At last, we spread out the results and summarized the rules. In the third part, we established the database, which included both basic information and medicament composing, of formulas for amnesia. On the basis of the database platform, we took information in the formulas by bibliometric, statistic and data-mining method including Apriori arithmetic and so on. Otherwise, we depicted and analyzed materials in database. In the fourth part, we collected 50 patients with MCI from communities and clinic. The patients were randomly divided into treatment group (33 cases) with 5 pieces Shenwu Capsule given orally three times daily three months or control group ( 17 cases) with 1 tablet donepezil once daily three months. At the same time, the symptoms of MCI were observed and analyzed. At the end of the therapy, the efficacy of the drug was monitored by the rating scales of Chinese medicine syndrome and neuropsychological tests. Besides, we briefly estimated the security of Shenwu Capsule for patients with MCI.Results: We summarized the characteristic of the Chinese medicine that is used to prevent and cure amnesia from ancient herb literatures as follows:(1)Drug has mainly the nature of sweet and equal, less cold or hot, and much is also pungent or bitter(;2)Drug is mainly distributed in the upper grade or middle grade in the ancient herb literatures, and Junyao has larger proportion;(3)Drug is mainly plant, and smooth and slow with less toxicity;(4)Syndrome differeation mainly based on the heart and the kidney. The basic principles for making syndrome differentiation and curing amnesia in Chinese medicine were generalized as follows:(1)Syndrome differentiation mainly based on the heart,the spleen and the kidney, and gave attention to other visceras.(2)Attach importance to the surplus or deficiency of the basic substance such as essential, Qi, blood, marrow and so on;(3)The principle of treatment is mainly complementing the deficiency and combining complementing the deficiency to allaying Xie.(4) The methods of treatment are tonifying the deficiency, opening the orifices, soothing the nerves, warming the inner and activating the blood and so on. The drug for amnesia should mainly have the nature of equal or warm, and sweet, pungent, bitter. In the study, we have got the 10 materials used frequently for treating amnesia in Chinese medicine, such as Radix Ginseng, Radix Polygalae, Poria, Radix Glycyrrhizae, Poria inner, Radix Angelicae Sinensis, Radix Ophiopogonis, Cinnabaris, Rhizoma Acori Tatarinowii, Ramulus Cinnamomi. We also have aquired Yaodui, such as Rhizoma Anemarrhenae matching Cortex Phellodendri, Bombyx Batryticatus matching Scorpio and so on, by association rule mining. In the clinic research, the total effective rate was 58.06% in treating group, and 58.82% in control group. It also showed Shenwu Capsule could significantly improve such symptoms as weakness and soreness in the low back and knees, lassitude, feeling of heavy head, headache like stinging, tinnitus, swirling and so on (P < 0.01). Statistical analysis evidenced significant improvements in cognition in the treatment group in respect to the control group, immediate story recall and delayed story recall(P=0.000), Instrumental Activities of Daily Living Scale (IADL) (P<0.01), Alzheimer's Disease Assessment Scale- Cognitive (ADAS-cog) (P=0.000). Further, the patients in the treatment group could be improved in words recalling and identifying, attention and so on(P<0.01). However, the performing ability of the patients in the control group could also be significantly improved. (P=0.009).Conclusions: The prevention and cure of MCI can use the results acquired from studying Chinese medicine literatures for reference. Shenwu Capsules made at the guidance of the principle of strengthening spleen and benefiting kidney, dispelling sputum and opening orifice,activating blood and dispelling blood stasis have efficiency on patients with MCI with the syndrome of weakness both spleen and kidney mixed sputum and blood stasis. Shenwu Capsule also could markedly improve the patients'cognitive function, such as story scenario recalling, the ability of using instruments, the level of general cognitive function, especially words recalling and identifying, and attention, but had poor influence on the high central frontal function. |
