The adoption of this study was tostudy the relationship between heart and kidney, strengthen the heart and kidneydisease study of mutual ties and gradually establish integration, including theprevention of heart and kidney integration therapy, the best treatment for patients,improve the quality of patient's life and prognosis. This study from the followingtwo aspects of heart and kidney disease and the restoration of viscera in theprocess of interrelated preliminary exploration, in order to further provide thebasis for indepth study.Part 1: Established rat model of Hyperlipidemia combinedMyocardial ischemia and testing the function of heart and kidney.Objective: To established rat model of Hyperlipidemia combined Myocardialischemia and observe under pathological changes in the function of heart andkidney conditions.Methods: Prepared rat model of Hyperlipidemia combinedMyocardial ischemia by feeding rats with high lipid food and ligating coronaryartery which reference to literature methods and upgrading, and determination ofdifferent time of the animal models'ECG, blood biochemical,urinary biochemicaland pathological morphology, and other indicators by means of different groups ofanimal model, according to documents and experimental conditions specific toevaluate the function of heart and kidney. Results: 1.The levels of rats'totalcholesterol in serum increased. 2.Model rats showed myocardial lesion andabnormity of cardiovascular function: The positive rate of ischemia ECG in ratsincreased , tne levels of myocardial injury related enzymes in serum increased.Pathological Results: Part of the left ventricular myocardium was gray, endoscopicHE staining showed that regional myocardial tissue degeneration and necrosis, thelesion muscle fibers arranged disorder, muscle wire fault dissolution, stripesunclear, condensation nuclei, nuclearair bubble formation, muscle fiber cellulosebetween exudative, fibroblast cells and capillary sprouting, and inflammatory cellinfiltration. 3. Model rats showed abnormity of renal function: Creatinineclearance rate (Ccr) decline. Pathological Results: Glomerular mesangial showfocal segmental increase, the increase in cell number and a small amount of mild inflammatory cell infiltration, and the focal segmental expansion of tubular.Conclusion: dysfunction and tissue lesion of heart and kidney have been detectedin the rat model of hyperlipidemia combined myocardial ischemia simultaneously.Part 2: Observed the effects of methods of huoxuehuayu includingpharmacological effects and recovery effects of the function ofheart and kidney the.Objective: To observe the effect of Blood Circulation injection inShuxuetong injection (the group) and Danshen injection (single agent). Methods:Testing ECG, blood biochemistry, urinary biochemical and pathological changesin morphology, and other indicators of the animal model under the treatment ofDanshen high dose (20 mg/100g weight), medium dose (4 mg/100g weight) andShuxuetong high dose(0.3 ml/100g weight), medium dose (0.06 ml/100gweight).Results: 1.The positive rate of ischemic ECG and the levels of creatinekinase (CK) in serum decreased after the treatment of huoxuehuayu method.2.Creatinine clearance rate ( Ccr) was higher, urinary total protein (TP) and urineglobulin (GLO) was lower in treatment group than model group. Conclusion:Huoxuehuayu method can reduce the ischemia state of rat model of the coronaryheart disease and control early renal damage.To sum up: The rat model ofcoronary heart disease show myocardial ischemia and early renal damage, Therapyby Huoxuehuayu method can improve the pathological state of heart and kidney,and provide tne animal experimental demonstration for the initial realization oftreatment of heart and kidney simultaneously. |
