Shenmai Granula (SMG) is a compound recipe which is composed of six Chinese medicinal herbs including Radix Ginseng, Raidix Ophiopogonis and Radix Astragali.The effect of SMG with different components on decreasing blood glucose was investigated in diabetic mice induced by alloxan (ALX). The results showed that both crude extract of SMG and purified extract of SMG had significant effect on decreasing plasma glucose in diabetic mice after treatment for 14 days.Purified extract of SMG was selected to the next experiment according to the results in diabetic mice and the SMG product was made from purified extract.The effect of SMG on decreasing blood glucose and the antidiabetic mechanism of SMG was investigated in type 2 diabetic rats. Wistar rats were housed with high fat feeding for 13 weeks to induce insulin resistant rats and then treated with streptozotocin (STZ) to induce type 2 diabetic rats.The results showed that SMG (0.7, 1.4 g/kg, ig) had significant effects on antagonizing the decline of body weight, the growth of food intake and the increase of water intake, decreasing the level of plasma glucose, GHb, TG, T-CHO and FFA, inhibiting damage to oral glucose tolerance level, antagonizing insulin resistance, controlling hyperinsulinemia, decreasing the serum MDA level and increasing the serum NO level in type 2 diabetic rats which were treated with SMG for 28 days.The results in our study implied that the effect of SMG on decreasing blood glucose was associated with promoting the usage on glucose in tissue, stabilizing fluctuation of blood glucose, repairing and protecting pancreasβ-cell as well as increasing insulin sensitivity in type 2 diabetic rats. Furthermore, SMG also had a strong effect on decreasing blood lipid, agonizing lipid metabolic disturbance, eliminating free radical reaction and inhibiting oxidative stress. Hence, SMG could prevent the pathogenesis of diabetic complications.
