Objective:"Traditional Chinese medicine is a great treasure", and treatise on febrile disease, as one of the most famous representatives of four great classics of traditional Chinese medicine, is the gold room of the vast medical treasure-house.This dissertation intends to choose the classical prescription-huang lian e jiao soup as basic prescription, trough proper adding and subtracting, based on the thought of clinical return to the foundation, using science and technology, uniting modern medicine, successful inducing the type 2 diabetic rats model, evaluation hypoglycemic effect of add and subtract huang lian e jiao decoction on type 2 diabetic rats model, to test its hypoglycemic mechanism, simultaneous determination fasting blood glucose, fasting serum insulin, blood lipid and the changes of c-reactive protein of before and after the experiment, exploring the others effect of this soup, researching it’s effect on the sugar, islet B cell function, lipid metabolism and low-grade inflammation exist in the whole process of type 2 diabetes.To contribute to carry forward the charm of classical prescription, contributing to the modernization of traditional Chinese medicine.Methods:Through the methods of modern science, using the animal experiments confirmed that add and subtract huang lian e jiao decoction for type 2 diabetes mellitus hypoglycemic effect and other effects..Specific methods and process are as follows:phase one, choosing 40 healthy and lively male SD rats, adaptive feeding 7 days, based on weight, random selecting 8 rats for blank group,32 rats for model group, blank group was given common feed, model group was given high fat emulsion + common feed, feed cycle is 35 days.Phase 2, all rats fasting at least 12 hour and freely drinking water, model group rats were disposable intraperitoneal injected streptozotocin,30 mg/kg, blank group rats were injected same volume-lemon acid sodium citrate buffer, a week after shearing tail blood, using the blood glucose meter testing and recording fasting glucose, blood sugar more than 11.1mmol/L is as successful model.Phase three, after the success of the building, before drug intervention, collecting orbital blood, test blood lipid, fasting serum insulin, based on blood glucose and body weight, adopting stratified random methods 32 rats were divided groups, which is divided into four groups, namely model control group, model of low dose of Chinese medicine group, model of high dose of Chinese medicine group and model of Western medicine group.Phase four, drug intervention, model control group and the blank group, daily feeding amount of distilled water, high and low dose group model of traditional Chinese medicine to lavage and subtract huanglian ejiao decoction solution, the model of western medicine group to lavage metformin solution, at the same time all model group rats by gastric emulsion of high fat feed, gastric volume of 1 ml/100 g,1 times a day, the blank group continue to ordinary feed, cycle for 30 days.Stage 5, after the last delivery, fast water for 12 hours, with intraperitoneal injection of chloral hydrate anesthesia, big abdomen aortic blood collection, test three blood lipid, serum insulin, C reactive protein, the experiment ended, do statistical calculation, respectively do comparison and contrast between groups before and after drug intervention, assessment before and after drug intervention of blood glucose, serum insulin (insulin resistance), blood lipid, CRP changes. Insulin resistance index formula:HOMA-IR=FPG (tendency for L)×FINS (mU/L)/22.5, after calculating the natural logarithm.Results:1.FPG:compared with drug intervention before the state of sustained high blood sugar, high and low dose, add and subtract rhizoma coptidis gelatin soup for T2DM rats model glucose after intervention all have varying degrees of decline, with statistical difference (P<0.05); While high dose low dose showed better hypoglycemic effect (P<0.05),but western medicine hypoglycemic effect and does not show the glucose-lowering effectiveness is better than western medicine.2. The IR:after the success of the building, the model group compared with blank group rats showed obvious IR rats (P<0.05);IR to the Chinese and western medicine after the intervention, the intervention group was obviously improved (P<0.05), suggesting to add and subtract huanglian ejiao decoction has the effect of improving IR, high and low dose has no obvious differences in improving IR, improve the ability of IR has no obvious difference compared with western medicine.3.The blood fats:traditional Chinese medicine and western medicine on T2DM rats are regulation of blood lipids, can reduce the value of LDL (P<0.05), but didn’t show for TG, HDL, has the good has not shown between low dose and high dose of Chinese medicine and western medicine are different in terms of adjusting blood fat.4. CRP:CRP values of the model group were higher than that of blank group trend, but after statistical inspection, except the blank group and low dose group of Chinese medicine have differences, between groups did not show significant difference. This study did not confirm whether inflammation, is found in the T2DM rats model, and has not confirmed whether rhizoma coptidis gelatin soup have anti-inflammatory effects.Conclusion:Add and subtract coptis gelatin soup has a good effect on blood sugar, adjusting blood lipid disorders (mainly on the role of LDL) significantly and improve the IR;At the same time may have anti-inflammatory effects. The hypoglycemic mechanism should be related to improve insulin resistance.
