Hypoglycemic Effect Of Fermented Inonotus Obliquus On Type 2 Diabetic Rats

Objective:To explore the hypoglycemic effects of fermented Inonotus obliquus(FIO).on type 2 diabetic rats induced by Streptozotocin(STZ).Method:SD male rats(180+20 g)were randomly divided into normal group(NC)and model group(MC)according to body weight after one week of adaptive feeding.The rats of MC group were fed with fat emulsion for 15 days,and the contents of free fatty acids(FFA),total cholesterol(TC)and triglyceride(TG)in serum of the two group rats were detected.The rats of MC group were intraperitoneal(ip)injection STZ(30 mg/kg)for two times after the content of FFA in serum of rats were significantly increased,and the interval was one week.Second weeks after intraperitoneal injection of STZ,fasting blood glucose was measured in rats,and rats with fasting blood glucose greater than 7.8 mmol/L were selected as T2DM model;The T2DM rats were randomly divided into 4 groups according to their blood glucose,except for MC group,the other groups were given orally to FIO(1 g/kg),FIO(2 g/kg)and metformin hydrochloride(900mg/kg).After 8 weeks,The levels of FBG,FFA,TC,TG,high density lipoprotein(HDL),low density lipoprotein(LDL),malondialdehyde(MDA),superoxide dismutase(SOD),and insulin(INS)in serum of rats in each group were detected,and insulin sensitivity index(ISI)and insulin resistance index(Homa-IR)were calculated.The liver of rats in each group were collected and the content of glycogen in the liver was determined.The pancreatic of rats in each group were collected and stained with HE.The morphology of islet cells was observed under microscope.Results:1.Establishment of type 2 diabetes model1)After MC group rats were fed with fat emulsion for 15 days,compared with the NC group,the weight and daily food intake of rats were increased significantly in MC group(P<0.01),the levels of TC,TG and FFA in serum of rats were increased significantly in MC group(P<0.01).2)After the rats in MC group were injected with STZ for two times,compared with the NC group,the weight of rats was decreased significantly(P<0.01)and the level of FBG in serum of rats was increased significantly in MC group(P<0.01).2.Hypoglycemic effect of fermented Inonotus obliquus on type 2 diabetic rats1)Compared with the NC group,the level of FBG in serum was increased significantly(P<0.01),and the content of glycogen in liver was reduced significantly in rats of model group(P<0.01).Compared with the MC group,the content of hepatic glycogen was increased significantly in rats of FIO(1 g/kg)group(P<0.01);the level of FBG was decreased significantly,whereas the content of hepatic glycogen was increased significantly in rats of FIO(2 g/kg)group(P<0.01);Compared with FIO(1 g/kg)group,the level of FBG was decreased and the content of hepatic glycogen was increased in rats of FIO(2 g/kg)group(P<0.05).2)Compared with the NC group,the content of FFA in serum of rats was increased significantly in MC group(P<0.01).Compared with the MC group,the levels of FFA were significantly decreased in rats of FIO(1 g/kg)group and FIO(2 g/kg)group(P<0.01).3)Compared with the NC group,the contents of TC,TG in serum of rats were significantly increased in MC group(P<0.01).Compared with the MC group,the contents of TC,TG were significantly reduced in rats of FIO(1 g/kg)group and FIO(2 g/kg)group group(P<0.01).4)Compared with NC group,the level of HDL in serum of rats was decreased obviously,whereas the level of LDLin serum of rats was increased significantly in MC group(P<0.01).Compared with the MC group,the levels of HDL were increased obviously and the levels of LDL were significantly decreased in rats of FIO(1 g/kg)group and FIO(2 g/kg)group(P<0.01).5)Compared with the NC group,the content of MDA in serum of rats was increased significantly and the level of SOD in serum of rats was decreased significantly in MC group(P<0.01).Compared with the MC group,the contents of MDA were decreased significantly and the levels of SOD were significantly increased in rats of FIO(1 g/kg)group and FIO(2 g/kg)group(P<0.01);Compared with FIO(1 g/kg)group,the levels of SOD in serum of rats was increased in FIO(2 g/kg)group(P<0.05).6)Compared with the NC group,the level of INS was no change in serum of rats in MC group,the HOME-IR was increased significantly and the ISI was decreased significantly in MC group(P<0.01).Compared with the MC group,the HOME-IR was decreased(P<0.05),and the ISI was no change in rats of FIO(1g/kg)group,meanwhile the HOME-IR was decreased significantly(P<0.01),and the ISI was increased(P<0.05)in FIO(2 g/kg)group's rats;Compared with FIO(1 g/kg)group,the HOME-IR(g/kg)was decreased in FIO(2 g/kg)group's rats.7)Compared with the NC group,the structure of islet was destroyed,islet cells were scattered and uneven distribution,the number of islet ? cells were decreased significantly in MC group's rats;compared with the MC group,the structure of islet was tend to complete,the number of islets ? cells was increased in rats of FIO(1 g/kg)group and FIO(2 g/kg)group.In particular,the damaged pancreatic islet was recovered significantly,the number of ? cells was increased significantly in rats of FIO(2 g/kg)group.Conclusion:The FIO could effectively lower the blood glucose and plasma lipid,and improve antioxidant ability.
