| ObjectiveWith various modern research technique, this experiment is to study the effect of "shuang gu yi tong" acupuncture therapy on diabetes mellitus rats and its possible mechanism, and to compare the curative effect of"shuang gu yi tong" common stimulation acupuncture group and "shuang gu yi tong" strong stimulation acupuncture group.This will provide new method for acupuncture clinic and offer experimental evidences for the further research.MethodsDM models were made with Wistar rats by injecting streptozotocin into their lower left celiac cavity. Ten normal rats were divided into normal control group (group A).DM rats were randomly divided into four groups: model control group(group B), "shuang gu yi tong" common acupuncture group(group C) and "shuang gu yi tong" group with electroacupuncture of high intensity (group D). "guanyuan" and "zusanli" acupoints as the "shuang gu" acupoint, "yishu" as the "yi tong" acpoint. Group C and D are treated with G6805 acupuncture therapy- apparatus.Group C is treated with the intensity of 1 AM, lasting 15 minutes. Group D is treated with the intensity of 6 AM, lasting 30 second .The treatment lasted for two courses, 6 days in each course and two days at interval. Blood sugar was tested with sugar equipment. Serum insulin was measured with radioimmunoassy (RIA) and insulin sensitivity index was caculated accordingly. Expression of InsRβ(insulin receptorβ) and PI-3Kp 110 (Phosphatidylinositol 3-kinase p110) in the pancreas of the rats were detected by immunocyto- chemistry SABC method. Image analyse system was used to measure the immune positive InsRβ, PI-3Kp110.Results1 The therapeutic effect on the blood sugar of DM rats: In the treatment groups, the level of the blood sugar lowered after the treatment (P＜0.05). The effect of group D was better than that of group C. The insulin level of group C and D increased (P＜0.05). The two treatment groups could reduce water and food intake and increase weight of the DM rats, lower ISI obviously (P＜0.05). There was no statistic difference between the two groups ( P＞0.05 ) .2. Compared with the normal group, the expression of InsRβin the pancreas of DM rats decreased ( P＜0.05 ) .The expression of InsRβobviously increased in group C and D (P＜0.05) . There was no statistic difference between the two groups in increasing the expression of InsRβ( P＞0.05 ) .3. Compared with the normal group, the expression of PI-3Kp110 in the pancreas of DM rats decreased ( P＜0.05 ) .The expression of PI-3Kp110 significantly increased in group C and D ( P＜0.05 ). There was no statistic difference between the two groups in increasing the expression of PI-3Kp110 ( P＞0.05 ) .Conclusion1. " shuang gu yi tong " acupuncture therapy has reliable effect of down-regulating blood sugar on DM rats, up-regulating serum insulin and improving the abnormal metabolism and insulin resistance.2. "shuang gu yi tong" acupuncture therapy with different intensity increased the lower expression of InsRβ, PI-3Kp110, which may be impertant tache of "shuang gu yi tong" acupuncture therapy in regulating insulin signal ways and improving insulin resistance of DM rats.3. The curative effect between group C and group D has no significant difference. Group D can obviously reduce the treatment time, so it will provide new method for acupuncture practice.4.The above results provide scientific evidences for "shuang gu yi tong" acupuncture therapy in treating and preventing DM and its complication indicate that the therapy which emphasizes "protecting vital-qi" can regulate body function wholly and generally. |
