MethodsDM models were made with Wistar rats by injecting streptozotocin into their lower left celiac cavity. Ten normal rats were divided into normal control group (group A).DM rats were randomly divided into four groups: model control group(group B), "shuang gu yi tong" common acupuncture group(group C) and "shuang gu yi tong" group with electroacupuncture of high intensity (group D) . The treatment lasted for two courses, 6 days in each course and two days at interval. Blood sugar was tested with sugar equipment. Serum insulin was measured with radioimmunoassy (RIA) and insulin sensitivity index was caculated accordingly. Expression of InsRP (insulin receptor β) and PI-3Kp110 (Phosphatidylinositol 3-kinase p110) in the liver of the rats were detected by immunocytochemistry SABC method. Expressions of GLUT4(Glucose transporter-4) mRNA in the liver of the rats were detected by in situ hybridization. Image analyse system was used to measure the immune positive InsR|3, PI-3Kp110 and GLUT-4mRNA. Results1 The therapeutic effect on the blood sugar of DM rats: In the treatment groups, the level of the blood sugar lowered after the treatment (P<0.05). The effect of group D was better than that of group C. The insulin level of group C and D increased (P<.05). The two treatment groups could reduce water and food intake and increase weight of the DM rats, lower ISI obviously (P<0.05). There was no statistic difference between the two groups ( P>0.05 ) .2. Compared with the normal group, the expression of InsRP in the liver of DM rats decreased (P<0.05 ) .The expression of InsRP obviously increased in group C and D ( P<0.05 ). There was no statistic difference between the two groups in increasing the expression of InsRp (P>0.05 ) .3.. Compared with the normal group, the expression of PI-3K.p110 in the liver of DM rats decreased( P<0.05 ).The expression of PI-3Kpll0 significantly increased in group C and D (P<0.05 ) . There was no statistic difference between the two groups in increasing the expression of PI-3Kp110 ( P>0.05 ) .4. Results with in situ hybridization showed: Compared with the normal group, the expression of GLUT4mRNA in the liver of DM rats decreased ( P<0.05 ) .The expression of GLUT4mRNA significantly increased in group C and D (P<0.05 ) . There was no statistic difference between the two groups in increasing the expression of GLUT4 gene (P>0.05) .
