Objectives:To explore the association between the polymorphisms of transforming growth factor-β1(TGF-β1)gene and human longevity.Methods: Four polymorphisms located in the promoter region(G-800A,C-509T)and the exonl(T+869C,G+915C)of the TGF-β1 gene were genotyped with polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) technique in 132 Zhuang healthily long-living people and 118 Zhuang healthy controls from Bama area and 70 Zhuang healthy adults from Guangxi Hezhou area.Then the polymorphic genotype frequencies and allelic frequencies were analyzed among the three groups.Results:1.The polymorphisms of G-800A and G+915C on TGF-β1 gene were not found in all studied individuals.2.The polymorphisms of C-509T and T+869C were detectable,but no allelic frequency and genotypic frequency differences were found between the long-living group and the controls in Bama area(p>0.05).Nevertheless,when compared to Hezhou Zhuangs,both Bama long-living group and Bama controls showed significant differences.In detail,on -509 locus,the allelic and genotypic frequency of Bama long-living group and Bama control group were both greatly different from that of Hezhou Zhuangs(pallelic=0.000,0.034;pgenotypic=0.000, 0.039,respectively);on +869 locus,the allelic and genotypic frequency of both Bama groups were also apparently different from that of Hezhou Zhuangs (pallelic=0.000,0.017;pgenotypic=0.003,0.026,respectively).3.Further combining genotype analysis showed that,the frequency of genotype combination of GG-CT-TT-GG on G-800A,C-509T,T+869C,G+915C loci of Bama long-living group was significantly higher than that of Bama controls(p=0.000); the genotype combination of GG-CT-TC-GG and GG-TT-CC-GG of Bama long-living group and Bama controls were both greatly different from that of Hezhou Zhuangs(pGG-CT-TC-GG=0.002,0.002;pGG-TT-CC-GG=0.002,0.031, respectively).Conclusions:The allelic,genotypic and combined genotypic frequencies of some loci on TGF-β1 gene of Bama long-living population were significantly different from that of common cohorts and certain loci showed associations with longevity to some extent in Bama area,suggesting that Bama long-living people might have harbored unique anti-inflamm-aging mechanisms which contribute to their successful aging.
