TCM has a long drug detoxification history and also has been widely used in contemporary detoxification, especially in alleviating protracted abstinence symptom and reducing physiology dependence.But there is no syndrome differentiation and comparative neurobiology base in the protracted abstinence symptom of opium-addicts.It make the therapy cannot standard and objective which affluent development.We get the traditional Chinese medicine Syndromes of PAS through research illicit drug abusers in clinical survey. And select common syndromes to establish combined morphine dependent and Chinese medicine syndrome rat model which examine correlative neurobiology index. we reveal the difference of Neurobiology in different Chinese medicine syndrome.There are three aspects, the theoretical studies, clinical investment and animal experiment to research traditional Chinese medicine Syndromes and neurobiology machine of protracted abstinence symptom of opium-addicts.In theoretical studies, we discuss the TCM understanding and treatment to PAS through search academic monograph and journal. In clinical investment, we have a large sample test and get overall syndromes. In animal experiment, we establish the morphine dependence and traditional Chinese medicine syndrome combined rat model first and research the difference of Neurobiology in different Chinese medicine syndrome.Theoretical studiesIn this part, we generalize the TCM understanding and treatment to PAS through search ancient and contemporary academic document.1 TCM theory to the PASTCM consider that PAS has four classes symptom. One is deficiency symptom, two is psychological craving symptom. Three is pain symptom. Four is stomach intestine symptom.The pathogenesis of PAS is involved with qi-blood damaging, deficiency yin affecting yang, zang-fu deficiency.The syndrome of PAS is involved qi-blood deficiency, zang-fu deficiency, yin-yang deficiency, qi stagnation, blood stasis, phlegm stagnation etc.2 TCM treatment to the PASThe principle of detoxification in TCM is supporting healthy energy to eliminate evils, invigorating yang and nourishing yin, restoration zang-fu, dispelling Smoke Poisonous, Adjusting Activity of Qi-blood. Regulation is the main treatment in last stage.The Detoxification TCM Prescription divides into Emergency treatment Prescription, Detoxification Prescription, Regulation prescription. There were some ancient prescriptions and new drugs in contemporary age to cure PAS. In conclusion, TCM form systemic theory and clinic experience to PAS. As a characteristic treatment, TCM will gain the affirmation and development in the world.Clinical Studies1 An analysis for psychosocial factors of opioid addictsThis part analyzed 1030 illicit drug abusers , They were collected and research in demography and fundamental drug abuse condition. Illicit drug users tended to be young, less educated, having no stable occupation and formal marriage relationship. The age of initial use between 15 to 25.The main illicit abusing drug is heroin and the main mode of abusing drug is intravenous injection. Among them 61.70% abusers use the same injector with others.50.10% person mainly abuses place at home residence.There are 92.90%people has relapse and average relapse frequency is 4.81±s3.63 times,the average abstinence drug is 219.97±s304.70 days. The main reason of relapse is the influence of the mental factor.2 The symptoms and Traditional Chinese Medicine syndrome of PAS 1004 PAS opium-addicts were analyzed with self-designed'protracted abstinence symptom of opium-addicts Chinese medicine syndrome questionnaire'.The result indicates that the top 10 symptoms is fatigue, loin ache, sleepiness, Tired and speech less, forgettery, joint ache, insomnia, the muscle power decrease, sight decrease, extremity sweat, the symptoms coincidence rates are above 50%.Most symptoms of PAS are related to lung, spleen, liver, heart, and kidney, the top 10 Traditional Chinese Medicine syndrome of PAS is kidney deficiency, qi deficiency lung deficiency, spleen deficiency, blood stasis, phlegm dampness, dampness heat, heart deficiency, liver deficiency, excess heat.The main symptoms is deficiency disease.3 The psychological characteristics of detoxification addicts and recovery treatmentSCL-90 scale was used to test and compare two groups of detoxification addicts. One group take part in the'ten step'recovery treatment and other group do not. The results of two groups were finally compared with the normal control. The recovery treatment includes cognitive psychotherapy, behavioral therapy family treatment and social skill training. The results is except hostility, other factorial scores including positive items, somatization, obsession, interpersonal sensitivity, depression, phobia, paranoid and psychotic factors among them who do not attend recovery treatment significantly higher than the people who take part in the recovery treatment.Experimental Studies1 Establishment and evaluation of combined Syndrome and Disease rat modelBased on massive clinical study to establish the morphine dependence and traditional Chinese medicine syndrome combined rat model, we collected qi deficiency and qi stagnation two common traditional Chinese medicine syndrome in PAS to establish. First we built and evaluated syndrome rat model by traditional Chinese medicine theory, then stacked morphine dependence model to establish the combined syndrome and disease rat model in succeed.2 The research on neurotransmitters in different Syndrome of morphine dependence ratTo test DA, 5-HT, Glu, cAMP, cGMP in normal group, morphine dependence group, qi deficiency+morphine dependence group and qi stagnation+ morphine dependence group. Results indicate that:(1)Morphine dependence group, deficiency+morphine dependence group and qi stagnation+morphine dependence group have significant difference in comparison with normal group.(2)Hippocampus DA, hippocampus cAMP and cortex cAMP of qi deficiency+morphine dependence group and qi stagnation+ morphine dependence group have significant difference in comparison with morphine dependence group.(3)The qi stagnation+morphine dependence group's content of DA, Glu, CGMP in hippocampus are higher than qi deficiency+morphine dependence group's, and the result is opposite in cortex.(4)there are significant difference between hippocampus and cortex in cAMP of Morphine dependence group and deficiency+morphine dependence group.
