Objective: To study the relationship between the expression of hypoxia inducible factor-1α and tumour angiogenesis in lung cancer.Methods: We selected 100 cases of surgical specimens of lung cancer. Of these patients , 58 cases were men, 42 were women, age ranging from 39 to 71 years old , mean age at 65.2. Histological types: 45 cases were squamous cell carcinoma, 38 were adenocarcinoma , 11 were small cell carcinoma , 6 were large cell carcinoma . Lymphatic metastasis: 63 cases were positive, 37 were negative. TNM stage: 37 cases were I stage, 21 were II stage, 36 were III stage, 6 were IV stage. We built one lung cancer tissue chip which included 100 cases of lung cancer tissue and 20 cases of normal lung tissue. By using immunohistochemistry and in situ hybridization, the expression of HIF-1α , HIF-1α mRNA, VEGF and VEGFmRNA was detected. And CD34 mAb was used as a marker of vascular endothelial cell to calculate microvassel density(MVD).Results: The positive rates of HIF-1 α , HIF-1α mRNA, VEGF and VEGFmRNA in lung cancer tissue were apparently higher than in normal lung tissue (P<0.01). The expression of HIF-1α , HIF-1α mRNA, VEGF and VEGFmRNA had no relation with the histological type of lung cancer (P>0.05). The expression of HIF-1α , HIF-1α mRNA, VEGF and VEGFmRNA in positive lymph node metastasis group was significantly higher than in negative lymph node metastasis group (P<0.01), and the expression of HIF-1α , VEGF and VEGFmRNA of lung cancer in III-IV stage was significantly higher than that of lung cancer in I-II stage (P<0. 05, P<0. 01, P<0. 01), but there was no significant...
