Abjective:The effect observation of TaohongSiwu decoction solution on ischemic reperfusion injury of limbs in rats skeletal muscle VEGF expression and microvessel density, to explore the mechanism of TaohongSiwu Decoction in the treatment of limb ischemia reperfusion injury.Methods:66SD rats were randomly divided into control group (6rats), ischemia reperfusion group(IR group)(30rats) and Taohongsiwu liquid group(group T-IR)(30rats). The control group without blocking the blood flow, the last two groups make animal reproduction right hind limb ischemia reperfusion injury model. According to the time after reperfusion were respectively divided into12h,1d,3d,5d,7d after reperfusion(6rats for each time point).The control group feeds without drug, IR group and T-IR group rats were given saline and TaohongSiwu decoction gavage respectively by gavage once12hours before ischemia,after ischemia and after reperfusion. Then gavage once daily in the morning and evening. Rats in each group at the corresponding time point make materials producer, HE staining was used to observe the morphological changes of skeletal muscle cells, VEGF and microvessel density was detected by immunohistochemistry.Results:(1)The results of HE staining:The control group of skeletal muscle cell shape and arrangement of the rules, uniform dyeing. The T-IR group and the IR group appeared different degree of muscle cells arranged in irregular, edema, muscle cell gap widened, destruction at each time point, but the IR group was more obvious.(2) The results of VEGF analysis:IR group and T-IR group VEGF early rising at each time point, after reperfusion7d began to decline, compared with the control group were statistically significant at all time points (P<0.01), But the T-IR group VEGF were higher than IR group at each time point, there was statistical significance at each time point except after reperfusion12h.(3)The results of skeletal muscle capillary density:The number of CD34positive cells was lower than that of the control group after reperfusion24h,the difference was not statistically significant (P>0.05),the other time points were higher than that of the control group,the differences were statistically significant(P<0.01). The skeletal muscle vascular density of T-IR group early rising,after reperfusion7d began to decline, but higher than that of group IR, the difference was not statistically significant(12h P>0.05), other time points were statistically significant (P<0.01).Conclusion:TaohongSiwu decoction can enhanced the expression of VEGF, increase the number of skeletal muscle capillary number in skeletal muscle of limb ischemia reperfusion injury, it mechanism of treatment of LIRI probably by enhancing the expression of VEGF in skeletal muscle after LIRI in rats, and promote angiogenesis, accelerate microcirculation reconstruction,thus reducing the severity of LIRI.
