Objective To investigate the genetic polymorphisms of SNP markers on Y chromosome in Shanghai,Guangdong and Xinjiang Chinese Han population,and evaluate their values in forensic application.Material and Methods The 71 Y-SNP polymorphic markers in Eastasia population has been choose by review related literatures and bioinformatics databases.Blood samples were collected from 205 unrelated males and 201 unrelated males of the Han population sample from Shanghai and Guangdong separately,saliva samples were collected from 201 unrelated males of the Han population sample from Xinjiang China.Genotyping of 71 markers was carried out in the samples by using multiplex PCR and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry.Allele frequencies of 71 Y-SNP markers were calculated by direct counting,the gene diversity(GD)and haplotypes diversity(HD)were calculated by formulas,the haplotypes frequency and population genetics comparison were collected and calculated by Arlequin version 3.5.2.2 software.Results In the Han population from Shanghai,allele frequencies of 66 Y-SNP markers were collected.The 62 of 66 Y-chromosomal SNP markers was informative,15 markers have high polymorphisms(GD ? 0.3),31 markers have medium polymorphisms(0.2 ? GD<0.3),16 markers have low polymorphisms(0.01<GD<0.2).GD were from 0.0098 to 0.5019,85 haplotypes has been found,HD was 0.9770.In the Han population from Guangdong,allele frequencies of 67 Y-SNP markers were collected.The 63 of 67 Y-chromosomal SNP markers was informative,11 markers have high polymorphisms(GD ? 0.3),16 markers have medium polymorphisms(0.2 ? GD<0.3),36 markers have low polymorphisms(0.01<GD<0.2).GD were from 0.0191 to 0.4956,67 haplotypes has been found,HD was 0.9632.In the Han population from Xinjiang,allele frequencies of 67 Y-SNP markers were collected.The 63 of 67 Y-chromosomal SNP markers was informative,23 markers have high polymorphisms(GD ? 0.3),21 markers have medium polymorphisms(0.2 ? GD<0.3),19 markers have low polymorphisms(0.01<GD<0.2).GD were from 0.0100 to 0.5022,170 haplotypes has been found,HD was 0.9982.The differences among the Shanghai Han,Xinjiang Han and Guangdong Han were statistically significance.Conclusion Genetics data of the Shanghai Han,Xinjiang Han and Guangdong Han has been collected,which proved the MALDI-TOF MS was able to type Y-SNP markers.The most of 71 Y-SNP markers and the system was informative and valuable in kinship testing and individual identification.
