Puerarin-eluting Stents On The Efficacy Of The Rabbit Iliac Artery In-stent Stenosis And Preliminary Mechanism

Backgroud and Objectives:Implantation of stent is one of the main thepary of CHD(coronary heart disease) at present.Restenosis has been a tough problem in the field of cardiopathy. Drug-eluting stent can low the rate of restenosis effectively,and is a breakthrough to the prevention and cure of the restenosis appearing in the coronary artery interventional therapy. Rapamycin-eluting stent and paclitaxel-eluting stent are used frequently in clinic .There are many problems that drug -eluting stent faces with,such as long-term effect and high price。It is confirmed by previous study that Puerarin can facilitate VSMC .Puerarin can promote apoptosis of VSMC significantly , inhibit the proliferation of VSMC and Intimal Area(IA),pretect endothelial cell,restrain platelet aggregation and antiinflammate ,etc.We chose Puerarin-eluting stent which is relative cheap and widespread in our experiments.We compare the bare stent with rapamycin-eluting stent(FirebirdTM)made in China.,and observe the curative effect of Puerarin after the implantation of stent into the arteria iliaca of the rabbit in order to make clear that whether Puerarin -eluting stent could restrain the proliferation of VSMC and the happening of restenosis ;and whether Puerarin could restrain the proliferation and migration of VSMC in out-of-body experiment.Methods:This study include three parts:1. The effect of Puerarin to the migration VSMC and MMP-9,MMP-2.(1)The isolating culture and identity of arteriae aorta VSMC;(2)determine the effect of Puerarin to the proliferation and activity of VSMC by MTT chromatometry;(3)determine the effect of Puerarin to VSMC migration by Transwell cultivate product;(4)dedtermine the effect of Puerarin to MMP-9,MMP-2(migration-relate protain) by Elisa and gelatinum zymography。2.
