| Objective Graves'diease(GD) is a kind of normal autoimmune thyroid disorder.In the diease,the GD itself and Antithyroid drug(ATD) all can cause leukopenia,even agranulocytosis which can cause the infection ,septicemia,shock,e-ven thyroid storm .The former experiment has already confirm that Tablet-FFJK not only can lower the thyroid gland function effectively and control its symptom,but also prevent and cure GD with leukopenia to have the certain advantage. Through observing the change of the number of White Blood Cell,the nucleated cell of marrow,the GM-CSF and the Lysozyme,this research prescribe the mechanisms of Tablet-FFJK,providing the basis with the medicine and further developments for the clinic.Methods1 The model was reconstructed:The Wistar rats were mainlined 0.1ml,0.2ml,0.3ml,0.4ml,0.5ml Y.E bacterium water(5 × 108/ml) in trail in first,fifth,tenth,fifteenth and twentieth day.The blood and thyroid gland were drawed in twenty-fifth day.The FT3,FT4 and TSH were measured and the histological changes were observed by light microscopes..2 In this experiment,we divided the 70 Wistar rats into Normal group,Model group,Tablet-FFJK high dosage group,Tablet-FFJK middle dosage group,Tablet-FFJK low dosage group,Traditional Chinese medicinal group,western medicinal group at random.These rats were cured one month.Their blood and thighbones were drawed.The FT3,FT4 were measured by RIA,the TSH was measured by Chemiluminesceneassay. The number of White Blood Cell and the nucleated cell of marrow were counted by normal regulation.The serum level of the GM-CSF was measured by ELISA. The serum level of Lysozyme was measured by Turbidimetry.Results1 In the model group,the serum levels of FT3,FT4 was higer than Normal group(P<0.01),the serum level of TSH was lower than Normal group(P<0.01).The results of common pathology study showed that the thyroid follicle epithelial cells appeared with pole shape,and the thyroid follicle shortened.2 The serum levels of FT3,FT4 in the Tablet-FFJK high,middle dosage group and the western medicinal group were obviously lower than the Model group(P< 0.01).The treatment of the Traditional Chinese medicinal group was weaker,and did not show the difference with the Model group(P>0.05).The serum level of TSH also got the similar results.3 The White Blood cell number in the three Tablet-FFJK group and the Traditional Chinese medicinal group had not difference with the Normal group (P>0.05) ,however, it in the western medicinal group was much lower than the Normal group(P<0.01).4 The number of the nucleated cell of marrow in the three Tablet-FFJK group and the western medicinal group had not difference with the Normal group(P>0.05). The number of the nucleated cell of marrow in the Traditional Chinese medicinal group was much higher than the Normal group(P<0.01).5 The propotion of granulocyte in the nucleated cell of marrow in the Tablet-FFJK middle dosage group , the western medicinal group and the Traditional Chinese medicinal group was higher than the Normal group(P<0.05).6 The serum level of GM-CSF in every group did not show the difference (P> 0.05) .7 The serum level of Lysozyme in the three Tablet-FFJK group and the Traditional Chinese medicinal group had not difference with the Normal group (P >0.05),however, it in the western medicinal group was much higher than the Normal group(P<0.001).Conclusion1 The GD model made by trail Vain injected with Y.E bacteriurm water have expressed the characteristic of the GD.It is useful that the model is used to study medicine and pathlology.2 Through comparing with TH ang TSH,it was confirmed that Tablet-FFJK has the effect of antithyroid.3 Form the both sides of born and breakage of the White Blood Cell,we comprehensivly evaluated that Tablet-FFJK can protect the GD rat's White Blood Cell.This research showed that Chinese herbal medicine in the Tablet-FFJK can increase The number of the nucleated cell of marrow, enhance The propotion of granulocyte in the the nucleated cell of marrow and preve... |
