Epilepsy is an ordinary mental disease, which usually has an effecton the viscera. Among the viscera, the spleen and the stomach, locatedin the zhongjiao(middle energizer), can regulate visceral functions andaffect the mental activities. Dysfunction of their transportation andtransformation can produce phlegm-pathogen, which can induce mentaldiseases. So we think the spleen and the stomach, dominating psychic andmental activities, are important viscera and put forth the opinion oftreating psychic and mental disorders by mainly regulating the spleen andthe stomach. As one of the spleen-stomach-regulating prescriptions,Add-Subtract Tsaoko Anemarrhenae Decoction is applied to treat epilepsy,which is clinically and experimentally testified to have the function ofcontrolling several types of epileptic paroxysms and antagonize severalkinds of epileptic animal models. In course of antiepileptic activities,the prescription can change the content, potency and gene expression ofamino acid and neuropeptide neurotransmitters or neurotransmediators andtheir receptors in the animal brains, especially glutamate and itsN-methyl-D-aspartate(NMDA)receptor. It can synchronically reduce thecontent of the NMDA receptor and the expression of NMDAR1 mRNA, and canblock the formation of the kindling model like MK-801, a noncompetitiveNMDA receptor antagonist, but has no toxic and side effects MK-801 has,which suggests that the prescription has the possibility to be anantiepileptic drug acting on NMDA receptor system. However, it has so manycomponents that it isn't able to be further studied. Therefore, for thesake of its simplification, we has singled out and studied its componentsfor the first time, figured out its optimal compatible method and foundits main components.Theoretical Study: The author has collected and studied the records about epilepsy in thebook of Huangdi's Canon of Medicine, analyzed and discussed the etiology,pathogenesis, prevention and treatment of epilepsy recognized in that bookand its theoretical influence on the next generations. Subsequently, the author has primarily studied some knowledge ofepilepsy presented by later generations, mainly including epilepticclassification, etiology, pathogenesis, analyzed the relationshipsbetween the functional activities of the heart, liver, kidney and spleenVI 草果知母汤加减抗癫痫作用的拆方研究and epileptic paroxysm, and simply illustrated correspondingtherapeutical methods and prescriptions for clinical reference.Experimental Study: Dividing the compound prescription of Add-Subtract TsaokoAnemarrhenae Decoction into four groups, the author usedpentylenetetrazol(PTZ) kindling rats, adopted behavioral method,observed antiepileptic effects of it and its decomposed groups, andscreened out two effective groups. In the following procedure, to exploretheir antiepileptic mechanisms, such methods as amino acid autoanalyticalmethod, radioimmunoassay and in situ hybridization were applied, thechange of cerebral amino acid and neuropeptide neurotransmitters ortransmediators in the rats was studied and the alteration of GABAAR mRNA,NMDAR1 mRNA and GABA receptor expression was researched into. The results are as follows: 1.Add-Subtract Tsaoko Anemarrhenae Decoction, Group 1 and Group 2degraded convulsive grade in kindling rats very obviously. Among them,the compound prescription was the most effective. Group 3 and Group 4 hadno remarkable effect. It is concluded that the treatment of epilepsy by regulating the spleenand stomach is feasible, the traditional treatment of epilepsy bymodulating the heart and liver is reasonable. Because of the best effectof the compound prescription, it is considered scientifically andreasonably compatible. 2.Group 1 and Group 2 were singled out and their antiepilepticmechanism was studied with the compound prescription.
