Syndrome, as one of the characteristics of theory of Zang, instructsthe clinical diagnosis and treatment and show integrated , change ,individual conception. So it has been paid expansive attention in studyingof traditional Chinese medicine in recent years. As basic syndrome of liverdiseases of Chinese medicine is concerned, liver stagnation syndrome isone of common syndromes in clinical practice and highly valuable forprofound research. Some researches indicate that liver stagnation syndromeis likely to result from regulating function disorder of high nervouscentre induced by negative psychic stress in which pathological changesof multi-system including nerve, endocrine, circulation, digestion,immunity, feeling and motion etc are involved. As a complicated syndrome,it is not only liver disorder itself but also complex system of multipleZang-fu and organs of total body are involved in liver stagnation syndrome.However, only part of known gene expression products (receptor, enzyme,cytokine and peptides etc) change is disclosed so far in some aspects,essence of liver stagnation syndrome is far away from uncovering. Complexmanifestations of syndrome are likely to have some material basis that ismost likely to originate from gene modulating disorder to its core. Asabove-noted knowledge is concerned, the following aspects were primarilyexplored in this thesis. 1.Theoretical research As the advent of era of post-genome, focal point of studying in lifescience has been transformed from structure genome to function genome. Thisdoctrine that pays more attention to gene function and gives more emphasison interaction and integration is in accordance with academiccharacteristics of Traditional Chinese Medicine. As syndrome research isconsidered as importance and crucial issue of development of traditionalChinese medicine, presenting the notion of syndrome genome is urgentlyneeded to cater for this chance and tendency. Syndrome genomics is thatmethod of function genome is applied to discover all the genes and theirfunctions related with some syndrome and make clear its essence in totalgene expression through exploring the gene variation and differentialexpression of syndrome in which same disease is divided into differentsyndromes and different diseases are classified as same syndrome whentheory of syndrome is taken as leading principle. Therefore, presenting the notion of syndromegenome is necessary and inevitable for profound advancement of subject ofsyndrome in Chinese medicine. 2 Experimental research 2.1 The imitation of Liver stagnation syndrome. Based on the foregoingresult, we use the model of chronic unpredictable mild stress to imitatethe Liver stagnation syndrome, and test the wastage of 1% saccharosesolution and the content of rat's blood plasma ACTH. We use male SD ratsand randomly divide into the normal control group which 10 rats was putin the same cage, the group of stress model and the group of stress modeladministered with amitriptyline which each rat was fostered solely, thefourth group was model group administered with Xiaoyaosan and each rat wasfostered solely. At the first day and the last day in experiment, we testthe wastage of 1% saccharose solution, and in the last day, by theanaesthesia, we get 2ml rat's blood in order to gain blood plasma. The resultshowed that the wastage of 1% saccharose solution of the normal controlgroup is more than that of the group of stress model, and was familiar withthe result of the content of ACTH.
