Objective:To obtain the changes of rats with the stagnation of liver-qi syndrome and the effects of electroacupucnture(EA) in treating the stagnation of liver-qi, we observed rats with stagnagnat-ion of liver-qi syndrome caused by chronic stress in three aspects including behavior, cerebral functional imaging and hormones related to hypothalamus-pituitary-adrenal (HPA) axis.The focus of this study is to obtain objective visibility basis of changes in brain function following the stagnation of liver-qi syndrome by using positron emission computed tomography (PET) technology, as well as revealing the central mechanism of EA in treating the damage of brain function following the stagnation of Liver-Qi syndrome.Methods:1 We adopt methods of chronic stress, combining with isolated raised to model rats with the stagnation of liver-qi syndrome.2 The rats were randomly divided into three groups according to the weight:the normal group, model group, EA group, n=10.3 We judge the behavioral changes of model rats and the behavioral action of EA in treating the stagnation of liver-qi syndrome through Open-Field Test, partial addicted to sugar water te-st, measurement of body weight, food intake and observation of general condition such as mental status, fur, intelligence, spontaneous activity and stool.4 Cerebral functional imaging experiment of model group and EA group rats were carried out in E-plus animal PET scanner with 18F-FDG as a tracer.This experiment adopt matching method. SPM software which is the gold standard in the field of brain image is adoped to do image analysis and statistical study. We obtain brain regions with different FDG metabolism that is brain activation maps through two-sample t-test of datas between model group (before modeling) and model group (after modeling), model group (after modeling) and EA group (after modeling),EA group (before modeling) and EA group (after modeling). Therefore, we obtain objective visibility basis of changes in brain function following the stagnation of liver-qi syndrome, as well as revealing the central mechanism of EA in treating damage of brain function following the stagnation of liver-qi syndrome. 5 We judge HPA axis state of rats with the stagnation of liver-qi syndrome and the effects of EA to HPA axis by the content of adrenocorticotropic hormone-releasing hormone (CRH) and serum corticosterone (CORT) that are measured by ELISA method.Results:1 Results of behavioral studies:After chronic stress procedure of 4 weeks, horizontal moveme-nt scores and vertical movement scores of rats in model group both significantly reduced compared with the normal group (P<0.01). Sugar consumption.the degree of partial addicted to sugar water and food intake of model group rats decreased significantly (P<0.01). as well as weight growth slower(.P<0.01). The indicators above all improved after acupuncture treatment (P<0.05 or P<0.01).2 Results of PET studies:①SPM analysis of related brain areas between model group rats (before modeling)and model group rats(after modeling):The related brain areas with abnormal glucose metabolism of in model group rats are very many. After 28d modeling, The brain areas with dcreased glucose metabolism included (P<0.001):left cerebral hemisphere:posterior central gyrus, anterior central gyrus; right cerebral hemisphere:anterior cingulate; bilateral cerebral hemispheres:posterior cingulate;left diencephalon:dorsomedial nucleus,lateral nucleu-s.The brain areas with increased glucose metabolism included (P<0.001):left cerebral hemis-phere:parahippocampal gyrus,middle temporal gyrus,superior frontal gyrus,long gyrus of insula;right cerebral hemisphere:striatum(gray matter); right diencephalon:ventromedial nucle-us,ventrolateral nucleus;left cerebellum;②SPM analysis of related brain areas between model group rats (after modeling) and EA group rats (after modeling):The brain areas of EA group rats with dcreased glucose metabolism included(P<0.001):left cerebral hemisphere:superior frontal gyrus,inferior frontal gyrus,superior parietal lobule;paracentral gyrus;supracallosal gyr-us;left diencephalon:zona incerta inferior nucleus,lateral nucleus;The brain areas with increase-d glucose metabolism included(P<0.001):left cerebral hemisphere:anterior central gyrus, pos-terior central gyrus, middle temporal gyrus,orbital gyri,striatum(gray matter);right cerebral hemisphere:superior frontal gyrus,anterior central gyrus; right diencephalon:subthalamic nucleus;③SPM analysis of related brain areas between EA group rats (before modeling) and EA group rats (after modeling):After 28d experiment, the brain glucose metabolism in EA group rats (before modeling) has no statistically difference compared with EA group rats (before modeling) within the setting threshold(P<0.001).3 Results of hormones related to HPA axis studies:The contents of CRH and CORT in serum of model group rats were significantly higher compared with the normal group (P<0.01);The contents of CRH and CORT in serum of EA group rats were significantly lower compared with the model group because of EA treatment (P<0.01).Conclusions:1 Rats with the stagnation of liver-qi syndrome caused by chronic stress have behavioral changes including lack of pleasure, decreasing of exploration activities, slower weight growth-ing, reducing food intake and others,which are consistented with clinical symptoms characteris-tics of the stagnation of liver-qi syndrome. EA can better the symptoms liver-qi stagnation significantly.2 The stagnation of liver-qi syndrome has abnormal glucose metabolism in related brain areas, and the glucose metabolism is closely related to each function, by which we obtain objective visibility basis of changes in brain function following the stagnation of liver-qi syndrome. EA can significantly better the abnormal glucose metabolism of related brain areas. Overall,it plays an increasing role in brain areas with decreased glucose metabolism,as well as playing a reducing role in brain areas with increased glucose metabolism.The result suggests that the action of two-way adjusting to abnormal glucose metabolism of related brain areas is one of the central mechanism of EA in treating the stagnation of liver-qi syndrome.3 The stagnation of liver-qi syndrome shows state of HPA axis hyperactivity, EA can inhibits the excessive secretion of CRH and CORT.The action of positive adjusting to HPA axis is one of the mechanisms of EA in treating the stagnation of liver-qi syndrome.
