Objective: To research the inhibiting effect of "Tongguan" capsule to the vasculogenesis of the hyperplastic prostatic tissue and it's principles in SD rats. Method: The castrated SD rats were applied to receive subcutaneous injection of testosterone in order to copy BPH model. Immunohistochemistry combined with Microimage Analysis System (MIAS) was adopted to study microvascular density (MVD) as well as the expression of vascular epithelium growth factor (VEGF) and it's receptor (Flk-1), base-fibroblast growth factor (bFGF) and endostatin in the prostatic tissue of every rat in each experimental group. Result: In the hyperplastic prostatic tissue, there was a distinct elevation of the MVD level and the positive expression of VEGF, Flk-1 and bFGF obviously elevated while the positive expression of endostatin lowered. Each group of different doses of "tongguan" capsule have a lower MVD level and VFGF, Flk-1, bFGF positive expression with higher endostatin positive expression than above. Conclusion: Using "tongguan" capsule to treat BPH and retract the volume of the prostate is in relation to it's inhibiting effect to the vasculogenesis of BPH. The principle of it's inhibiting effect to vasculogenesis is iu relation to the fact that it can inhibit the expression of vasculogenesis factor and promote that of vasculogenesis inhibiting factor of the hyperplastic prostatic tissue.
