Background and purpose:According to current n~dical research, metastase and recurrence are the wrost prognostic for breast cancer patients, therefore, early clinical diagnose and therapy play very important role for patients.Nuclear factor kappa B(NFkB),as a novel gene, can help to early diagnose for breast cancer patients. it expressed very low levels in healthy hurmn rrnim~ry glands of adult won~n. but it displayed significant levels in breast cancer patients? tissue. sc~ experts suggest that NEB ny be a novel mrker for the nmnageimnt of breast cancer. therefore, we examined NE-kB expression as a nm-ker for the detection of carciwm tissue of breast cancer and ccmared with clinical histopathological features to evaluate the clinical significance of NE-kB assay. Patient and nthods:65 fennle patients with priumy breast cancer were all accepted operation. 35 paients with nontumrigenic adjacent tissue,and 31 patients with benign disease of nmmary glands as control. the diagnose of all patients were identified by histopathological examination. samples procured in surgical remval of breast ti.imrs. then stored at . Total protein was extracted frw cancer tissue and control tissue, then we detected the expression of NE-kB by Western bloting technique and irrnunohistochemistry techniques. at the same time, we examined the expression of ER PR, PCN in 65 prinry tuinjrs by inrnunohistochemistry techniques. Results:The experiment shows that the expression of NE-kB is significantly high levels in 65 cases breast cancer tissue.but the expression of NE-kB is very low levels even absence in healthy memniry glands and benign breast disease, the expression of NE-kB in cancer tissue is not correlated with clinical stage,priimry tumr size, patients age, hoimne receptor status and PCNA. but correlated with axillary lyniphnodes metastases. Conclusions: This study suggested that:1. As a novel tumr marker, NE kB is sensitive for breast cancer. 2. As a transcription factors, activation of NE-kB plays an early, critical role in the carcinogenriven transfoimtion of rtuim~ry glands. 3. The expression of NE-kB in breast cancer tissue can be considered as an independence prognostic factor. 4. It helps to direct the clinical therapy after operation...
