Objective: To study the change of cyclooxygenase-2(COX-2) on the evolve -ement of breast cancer, and analysis its role and significance on carcinogenesis and evolvement of breast cancer, and its relations with ER,PR,HER-2 in breast cancer. Methods: The expression of COX-2 in normal breast tissues, benign breast tissues, atyp -ical hyperplasic tissues and breast cancer tissues was detected by immunohistochem -istry(IC) staining and was analyzed by microscopic scoring and computer picture -scanning analysis system. Results: The expression of COX-2 in breast cancer tissues, atypical hyperplasic tissues and benign breast tissues are higher than in normal breast tissues (high expression ratio:34%,20%,16%,0% P<0.0001,P<0.001,P<0.01).The expres -sion of COX-2 in breast cancer tissues is higher than in benign breast tissues (P<0.05).The expressions of COX-2 in normal breast tissues, benign breast tissues, atypical hyperplasic tissues and breast cancer tissues show a gradually higher trend and some linear correlation (r=0.332,P<0.001).The results of COX-2 expression in tissues by computer picture-scanning analysis system also show that the expression of COX-2 is highest in breast cancer tissues than in the atypical hyperplasic tissues, benign breast tissues and breast cancer tissues(integral photodensity: 110.722, 83.794,81.005,34.068). The expression of COX-2 in the positive groups(high expression ratio:35.7%,48%,50%) and the negative groups(high expression ratio:38%,28%,29%)of ER,PR and HER-2 do not show statistical difference (P>0.05).COX-2 is also highly expressed in negative groups of ER ,PR and HER-2 (positive ratio:88%,89%,86%).Conclusion:COX-2 plays an important role on carcinogenesis and evolvement of breast cancer. The breast atypical hyperplasia and benign tissues with higher expression of COX-2 maybe have higher risks to developing to malignant tissues than them with lower COX-2 expression. COX-2 maybe provide a new way to predict breast cancer risk .prevent and treat breast cancer.
