Objective Thyroid associated ophthalmo is an orbital autoimmune disease that is closelyassociated with Graves'hyperthyroidism, its pathogenesis remains unclear. The present studyhave shown that several cytokines (Cytokines, CK) such as interferon (Interferon, IFN),interleukin -1 (Interleukin -1, IL-1), IL-4, tumor necrosis factor (Tumor necrosis factor, TNF)and other involved in the occurrence and development of TAO. Nuclear factor-κB (NCR Clearfactor kappa B, NF-κB) is a protein factors with multifunctional transcription and regulatory role,it is compatible with a variety of cytokines and adhesion factor gene promoter binding sitesoccurring, and enhance these genes transcription and expression.we detect the expression ofNF-κB and TNF-αin the blood of TAO patients, to analyze the correlation with the TAO ,andprovide experimental basis for clinical treatment.Methods 160 subjects were divided into three groups,54 patients with TAO, 52 patientswith Graves disease(GD) and 54 normal controls. The serum levels of TNF-αare measured byEnzyme-linked immunosorbent assay (ELISA), the nuclear protein levels of NF-κB are measuredby Western blot.Results①TNF-αconcentrations were 6.157±0.653,14.088±2.598 and 2.398±0.326 inthe peripheral blood serum of TAO Group,GD group and the CON group;Compared with thecontrol group, the expression of TNF-αwas significantly higher in serum of TAO and GDgroups(P<0.01;P<0.01). TAO serum TNF-αconcentrations were significantly lower comparedwith GD group(P<0.01).②Nuclear protein NF-κB levels were 0.334±0.490,1.186±0.159 and0.077±0.0005 in peripheral blood of TAO Group, GD group and the CON group;Comparedwith the control group, the expression of NF-κB in peripheral blood of TAO and GD groups washigher(P<0.01;P<0.01). TAO peripheral blood NF-κB concentrations were significantly lowercompared with GD group(P<0.01).Couclusion①TNF-αand NF-κB expression was significantly increased in the peripheralblood of TAO and GD patients.②The high expression of TNF-αand NF-κB in the peripheralblood of TAO have no specificity.
