| Gastric ulcer (gastric ulcer, GU) is one of the common digestive disorders which occurs due to weakened gastric mucosal defense and increased mucosal aggravating factors. Gastric ulcer is mainly caused by Helicobacter pylori infection, non-steroidal anti-inflammatory drugs (NSAID),and excessive gastric acid secretion. This disease in Chinese medicine can be described and found in the area containing "Stomachache", "fullness", "noisy", "Tunsuan", "spitting blood", and "bloody stool. " The clinical manifestations mainly include abdominal pain, often accompanied by belching, heartburn, vomiting and loss of appetite. Complications are bleeding, perforation, pyloric obstruction, and stomach cancer."Stomach problem causes sleep disturbance, " In Chinese medicine, the relationship between the Stomach and sleep is already discussed in the "Yellow Emperor's Classic of Internal Medicine" which goes back two thousand years. According to "Suwon-nitiaolun" Yang Ming belongs to the Stomach channel; the Stomach is the Sea of six Fu(internal organs); the Stomach Qi flows downward and when it rebels, it flows upward. Thus, it is hard to sleep. Xia Jing Yue:the result is "stomach problem causes sleep disturbance." Thus, it is important to see the mechanism of the relationship between the Stomach and sleep during the occurrence of the disease."shenyongzhenzhe, yang is produced from yin, yang is formed from yin." The proper ways to use the needle are:harmonize yin and yang; open meridians and activate channels; control the flow of Qi. According to studies done, acupuncture works through the network of neuroendocrine-immune system and by regulating the neurotransmitters and hormonal secretion; thus, it increases the body's defense against stress-causing factors and prevents stress-related diseases. Acupuncture works through a multi-level, multi-channel, multi-adjustment means of control. Thus, it can have a protective effect on the gastric mucosal damage and a therapeutic effect on gastric ulcer.Purpose:Using an ethanol-induced rat gastric ulcer (alcoholic ulcer, AU) model, the study of acupuncture treatment, the sleep time of rats, the study of behavioral conditions, the study of morphological changes of gastric mucosa, and the level of hypothalamus octapeptide (cholecystokinin. CCK-8) are observed and investigated. The therapeutic effect of acupuncture on the AU and its impact on sleep are also observed. Then, the relationship between stomach ulcer and sleep is discussed and it provides some experimental evidence to "stomach problem causes sleep disturbance."Methods:Wistar male rats were randomly divided into 5 groups:a normal group, a model group, a treatment groupⅠ, a treatment groupⅡ, and a treatment groupⅢ, and each group contains 8 rats. The protocol of ethanol induced ulcer model is followed and used. The acupuncture point selections are Zhongwan, ZuSanLi, ZhaoHai, and Shen Mai. After treatment, locomotor activity of rats, the size of ulcer lesions, and ulcer index are calculated. Also, the sleep time and the level of CCK-8 in the hypothalamus are measured.Results:1. open field test:the model group was compared with the 4 groups and it was significantly different, P<0.01.2. ulcer index:when the model group and the treatment groupⅠand the treatment groupⅢwere compared, there was a significant difference, P <0.01; When the model group and the treated groupⅡwere compared, there was some difference, P<0.05.3. sleep time:when the model group and the normal group were compared, there was a significant difference, P<0.01; when the model group and all four treatment groups were compared, there was some difference, P<0.05.4. CCK-8 levels:when the normal group and the model group were compared, there was a significant difference, P<0.01; when the model group and all four treatment groups were compared, there was a significant difference, P <0.01.Conclusion:This study showed that:①acupuncture works by improving ethanol-induced rat's excitability②ulcer has some effect on the sleep time in rats, and after acupuncture treatments, the amount of rat's sleep time increased.③acupuncture can reduce AU rat's gastric mucosal injury, and promote mucosal repair.④acupuncture lowers the level of CCK-8 and by reducing the level, it possibly plays a role in the treatment of gastric ulcer.In summary, an acupuncture treatment has a therapeutic effect on AU, and when rat's gastric ulcer occurs, acupuncture has some effects on the sleep time. Also, based on the result that acupuncture can lower the level of CCK-8, it can possibly aid in the treatment of gastric ulcer. |
