Gastric carcinoma is the most common malignant tumor in China which derives from gastric mucosa cell.The clinical statistics show that gastric carcinoma was the first or second in the mortality of malignant tumor in our country.Because of lack of the specific symptoms and sings in early gastric carcinoma,while 5-year surgical survival rate in advanced gastric carcinoma is low and the prognosis is worse. Therefore,it is essential to explore the early gastric carcinoma diagnosis and the mechanism about invasion.In the field of tumor study in recent years,the study about cell apoptosis has become a focus in medical research.More and more datas also indicate that tumor is not only proliferation and differentiation abnormal,but also apoptosis abnormal.KISS-1 is a new tumor metastasis suppressor gene which was discovered in recent years.The expression of this gene can be detected in the organizations such as normal heart,brain,liver and lung.Gene mutation,loss and decreased expression of KISS-1 is related with the occurrence of many tumors.The mechanism of KISS-1 inhibite the metastas of tumor has not clarified.Some research indicate that maybe KISS-1 gene is by the way of accommodating the signal transduction access to inhibit tumor cells transferative ability,the activity of matrix metalloprotease and tumor cells proliferation.Ki-67,which is considered the ideal tumor marker to detect cell proliferation.It overlays each proliferative stage except Go.It saccurate function is still not clear currently.But by studying its expression and distribution in the cell nuclei,its function is considered to have the relationship with the chromatin and cells mitoschsis. So that it is the symbol of cell proliferation used most currently.The recent study shows that KISS-1 is not only related with tumor invasion, metastasis and proliferation,but also with induction of cell apoptosis.At present,there was no report about the research of the expression of KISS-1,Ki-67 in gastric carcinoma tissues and the relationship between their expressions with cell proliferation,apoptosis of gastric carcinoma.This study will detect the expression of KISS-1,Ki-67 protein in the 45 cases of gastric carcinoma tissues,23 cases of atypical hyperplasia tissues and 21 cases of normal mucosa tissues by the technique of immunohistochemical;at the same time to detect cell apoptosis in gastric carcinoma tissues by the way of TUNEL,in order to explore the relationship between them and cell apoptosis in gastric carcinoma.And then,seting forth their functions in the beginning and developing of gastric carcinoma.Materials and methods1.Using the technology of immunohistochemistry SP to detect the expressions of KISS-1,Ki-67 protein in the 45 cases of gastric carcinoma tissues,23 cases of atypical hyperplasia tissues and 21 cases of normal mucosa tissues.2.Using the technology of TUNEL to detect the apoptosis in the 45 cases of gastric carcinoma tissues,23 cases of atypical hyperplasia tissues and 21 cases of normal mucosa tissues.3.Statistical analysis:All the datas were analyzed by SPSS10.0 statistical package,the count information calculated the positive rate,the count information are analyzed by the Chi-square and spearman correlation analysis,enumeration date are expressed by standard deviation analyzed by the t-test and pearson correlationanalysis. The level of signficance difference isα=0.05.Results1.The expression of KISS-1,Ki-67 protein in different gastric tissues.1.1The experiment results show that KISS-1 protein positive signals in cytoplasm.The positive rates of the expression of it in normal mucosa tissues,atypical hyperplasia tissues and gastric carcinoma tissues is 90.48%(19/21),60.87%(14/23) and 35.56%(16/45) respectively.There is a significant difference between the three groups(P<0.05).1.2The experiment results show that Ki-67 protein positive signals in nuclei.The positive rates of the expression of it in normal mucosa tissues,atypical hyperplasia tissues and gastric carcinoma tissues is 14.28%(3/21),21.73%(5/23) and 48.88% (22/45) respectively.There is a significant difference between the three groups (P<0.05).2.The relationship between the expression of KISS-1,10-67 protein and gastric carcinoma differentiation.2.1The positive rates of expression of KISS-1 protein in gradeⅠ,ⅡandⅢof gastric carcinoma differentiation is 33.33%(7/21),36.36%(4/11)and 38.46%(5/13) respectively.There is no significant difference between them(P>0.05).2.2The positive rates of expression of Ki-67 protein in gradeⅠ,ⅡandⅢof gastric carcinoma differentiation is 33.33%(7/21),45.45%(5/11) and 76.92% (10/13) respectively.The positive rates of expression of Ki-67 protein increased gradually as the grade of differentiation of gastric carcinoma reducing.There is a significant difference between the three groups(P<0.05).3.The expression correlation analysis of KISS-1 and Ki-67 protein in gastric carcinoma.There is a negative correlation between the positive expression rates of KISS-1 and Ki-67 protein in gastric carcinoma tissues(r=-0.355,P<0.01).4.The comparison of apoptosis index(AI) in different gastric tissues.In gastric carcinoma tissues the average AI is(10.35±1.61%),but in the atypical hyperplasia tissues and normal mucosa tissues,there is no or several apoptotic cells,the average AI is(2.48±0.57)%,(1.00±0.19)%.The AI in the gastric carcinoma was significantly higher than that of atypical hyperplasiaand tissues and normal mucosa tissues,the AI comparison of each group had statistically significant(P<0.01). 5.The relationship of the expression of KISS-1,Ki-67 protein and AI in gastric carcinoma.5.1The average AI of KISS-1 protein expression in positive group and negative group in gastric carcinoma tissues are respectively(11.60±0.94)%,(9.66±1.48) %,the AI comparison between the two groups is remarkable significant.The expression of KISS-1 protein and AI had a positive correlation(r=0.586,P<0.05).5.2The average AI of Ki-67 protein expression in positive group and negative group in gastric carcinoma tissues are respectively(11.43±0.94)%,(9.32±1.42) %,the AI comparison between the two groups is remarkable significant.The expression of Ki-67 protein and AI had a positive correlation(r=0.666,P<0.05).Conclusion1.From normal mucosa tissues,atypical hyperplasia tissues,gastric carcinoma tissues, the positive rates of KISS-1 protein expression show a decreasing trend,which indicate that the decreased expression of KISS-1 protein may be correlated with gastric carcinoma.2.From normal mucosa tissues,atypical hyperplasia tissues,gastric carcinoma tissues, the positive rates of Ki-67 protein expression show an increasing trend.There is a negative correlations between the positive expression rates of Ki-67 protein and gastric carcinoma differentiation,which indicate that the increased expression of Ki-67 protein may be correlated with gastric carcinoma.3.There is a negative correlation between the positive expression rates of KISS-1 and Ki-67 protein in gastric carcinoma,which imply that the increased expression of KISS-1 protein can inhibite tumor cells proliferation.4.The expression of KISS-1 and Ki-67 protein has significant positive correlation with the AI in gastric carcinoma,which show that the expression of KISS-1 and Ki-67 protein may be correlated with the AI in gastric carcinoma.
