Objective: Malignant tumor has become a serious impact on human health and life ,currently,about seven million people each year die of cancer patients in the world ,and the number is increasing each year, in which the patients died of gastric carcinoma accounted for 700,000 cases. Gastric carcinoma as the most common cancer in gastrointestinal tumors,there are 934 thousand cases new cases each year worldwide, 42% (nearly 40 million) the number of new cases is in China, the situation has become very serious .But now the exact pathogenesis of gastric carcinoma is unclear ,with the widely application of molecular biology, molecular genetics of cancer have been gradually understood。At present, has confirmed that the occurrence of gastric carcinoma is more than a gene factor and multi-step complicated process。Kiss-1 gene is a tumor metastasis suppressor gene, studies has shown that it is related to the development and occurrence of many malignant tumors. It plays an important role in the process of invasion and metastasis. A variety of normal tissues, such as in the human heart, brain, liver, placenta, kidney, skeletal muscle and pancreas can be detected in the existence of kiss-1.β-catenin (β-cat) is an important part of the E-cadherin / catenin complex,it plays an important role in the maintenance of normal epithelial polarity and integrity. Its abnormal expression can makes the decline of adhesive force between cells and re-expression in metastasis, leading to the development of gastric cancer. As the important link of complex functions,β-cat cat not only mediateds adhesion between the same cells, but also can induces the nuclear transfer of Wnt signal and protocarcinogenic gene through Wnt/Frizzled receptor pathways ,at last induces the actiation of malignant change and cancer cells. In this experiment by detecting the expression of Kiss-1 andβ-catenin in gastric carcinoma,cancer adjacent tissues and normal gastric mucosa tissue , this paper discussed the relationship between the Kiss-1 andβ-catenin,and discussed the role of Kiss-1 andβ-catenin in the development and the relationship between clinicopathological characteristics,so as to provided the basis for diagnosis and treatment of gastric carcinoma.Methods: Immunohistochemical( streptavidin- peroxidase,SP) technique was applied to detected the expression of Kiss-1 andβ-catenin in 45 cases of gastric carcinoma,45 cases of the proximal adjacent tissues and 20 cases of normal gastric mucosa , and the comprehensive analysis of combined with the patient's age, gender, infiltration degree, lymph node metastasis clinicopathological factors and so on.Using the statistical software SPSS13.0 statistical data processing, count withχ~2 test, P < 0.05 is statistically significant. It analysed the relationship between the expression of both in gastric carcinoma by using Spearman rank corrdlationg which is one of non-parameter statistics. Reagents used: mouse anti-human Kiss-1 monoclonal antibody was purchased from SANTA CRUZ company,its working concentration is 1:400,rabbit anti-humanβ-catenin antibody was purchased from Bioword company,its working concentration is 1:150. SP immunohistochemistry kit was purchased from Beijing Boao Sen Corporation, DAB color reagent kit was purchased from Beijing Zhongshan Golden Bridge Biotechnology Co., Ltd..Results:1The expression of Kiss-1 andβ-catenin in gastric cancer tissues, adjacent tissues and control tissues1.1 Kiss-1 in normal gastric mucosa showed high expression,the positive rate was 90% (18/20),the positive expression of Kiss-1 in the proximal adjacent tissues was 64.44% (29/45) ,in gastric carcinoma was 48.89% (22/45).In normal gastric mucosa,the positive expression rate of kiss-1 was significantly higher than the proximal adjacent tissues the difference was significant (P <0.05),the expression in the proximall adjacent tissue was higher than normal mucosa,the difference was significant(P <0.05).1.2The expression rate ofβ-catenin in normal gastric mucosa was 100% (20/20),in the proximal adjacent normal tissue the expression rate was 46.67% (21/45),normally expressed in gastric cancer was 33.33% (15/45),β-catenin in the normal expression of normal gastric mucosa was significantly higher than that of the proximal adjacent tissues, the difference was significant (P <0.05), the proximal expression in cancer tissues is higher than the normal gastric tissue,there was a significant difference (P <0.05).2The relationship between the expression of both Kiss-1 andβ-catenin in gastric cancer and the clinicopathologic features of gastric cancer2.1The positive expression rate of kiss-1 in infiltrating serosa of the gastric cancer was 34.62% (9/26)),significantly lower than that of not invading the serosa,the positive expression is 68.42%(13/19). The positive expression rate of Kiss-1 in gastric cancer with lymph node metastasis was 35.71% (12/17) ,which was significantly lower than that without lymph node metastasis,the positive expression is 70.59% (10/28) (P <0.05). The positive expression of Kiss-1 had no significant relationship with patient's age, sex, degree of differentiation (P>0.05).2.2The positive expression ofβ- catenin in differentiated and well-differentiated gastric cancer was 38.70%(12/31),significantly higher than in poorly differentiated gastric cancer, the positive re-expression was 21.43% (3/14) (P<0.05), The positive expression rate ofβ- catenin in infiltrating serosa of the gastric cancer was was 46.15% (12/26),significantly higher than that of gastric cancer invading the serosa,the positive expression was 15.79% (3/19) (P <0.05). The positive expression rate ofβ-catenin in gastric cancer with lymph node metastasis was 17.86% (5/28),which was significantly lower than that without lymph node metastasis,the positive expression is 58.82% (10/17) (P <0.05). The positive expression ofβ-catenin had no significant relationship with patient's age,sex(P>0.05).3 The relationship between the expression of kiss-1 andβ-catenin in gastric carcinoma Using Spearman rank correlation analysis indicated that the Kiss-1 andβ-catenin expression in gastric carcinoma was positively correlated (rs = 0.502, P <0.05)Conclusions: (1) The expression of Kiss-1 in normal gastric mucosa showed high expression and it was closely related to the depth of invasion and lymph node metastasis; The expression of Kiss-1 in without serosal invasion in gastric cancer was significantly higher than in serosal invasion;The expression of Kiss-1 in gastric carcinoma with lymph nodes metastasis was significantly lower than in gastric cancer without lymph nodes metastasis (2)β-catenin was located in the membrane in normal gastric mucosa ,it was highly expressed and closely related to the degree of tumor differentiation, depth of invasion and lymph node metastasis.;The expression ofβ-catenin in differentiated and well-differentiated gastric cancer was higher than that in poorly differentiated gastric carcinoma; The expression ofβ-catenin in without serosal invasion in gastric cancer was significantly higher than in serosal invasion ;The expression ofβ-catenin in gastric carcinoma with lymph nodes metastasis was significantly lower than in gastric cancer without lymph nodes metastasis.(3)The expression of Kiss-1 andβ-catenin in gastric cancer was positively correlated both play an important role in the development of gastric cancer .Kiss-1 andβ-catenin may be closely related to the development,invasion,metastasis and prognosis of gastric carcinoma , the joint evaluation of their function has important clinical significance.
