Clinical Significance And Expression Of KiSS-1 Peptide Metastin And MMP-9 In Gastric Carcinoma

Objective: To investigate the expression of KiSS-1 peptide metastin and MMP-9 in gastric carcinoma tissues,gastric mucosa tissues from radical surgical margin and gastric mucosa tissues from surgical margin in patients with benign diseases to probe the clinical outcome of them and the correlation between metastin and MMP-9 in patients with gastric carcinoma.Methods: Immunohistochemistry of streptavidin-peroxidase(SP) method was used to detect the expression of KiSS-1 peptide metastin and MMP-9 in 65 cases of gastric carcinoma tissues, 65 cases of gastric mucosa tissues from radical surgical margin and 16 cases of gastric mucosa tissues from surgical margin in patients with benign diseases. The relationship between KiSS-1 peptide metastin and MMP-9 and their clinicopathologic parameters in patients with gastric carcinoma was analyzed. The correlation between metastin and MMP-9 were also analyzed.Results:1. Immunohistochemical staining revealed that the expression of KiSS-1 peptide metastin in gastric carcinoma tissues, gastric mucosa tissues from radical surgical margin and gastric mucosa tissues from surgical margin in patients with benign diseases was 35.38%, 73.85%, 87.50%, respectively. Expression of metastin was significantly lower in gastric carcinoma tissues than the level in gastric carcinoma tissues from radical surgical margin(P<0.05) and in gastric mucosa tissues from surgical margin in patients with benign diseases(P<0.05). No correlation was found between the expression of metastin with their pathological types in patients with gastric carcinoma. However, expression of metastin was correlated with clinical stage and lymph node metastasis, whichâ… -â…¡clinical stage(54.17%) was significantly higher thanâ…¢-â…£clinical stage(24.39%) and without lymph node metastasis(55.55%) was significantly higher than that with lymph node metastasis(27.66%), both of them have statistical significance(P<0.05).2. The expression of MMP-9 in gastric carcinoma tissues, gastric mucosa tissues from radical surgical margin and gastric mucosa tissues from surgical margin in patients with benign diseases was 73.85%, 29.23%, 25.0%, respectively. Expression of MMP-9 was significantly higher in gastric carcinoma tissues than the level in gastric carcinoma tissues from radical surgical margin(P<0.05) and in gastric mucosa tissues from surgical margin in patients with benign diseases(P<0.05). No correlation was found between the expression of MMP-9 with their pathological types in patients with gastric carcinoma. However, expression of MMP-9 was correlated with clinical stage and lymph node metastasis, whichâ… -â…¡clinical stage(58.33%) was much lower thanâ…¢-â…£clinical stage(82.93%) and without lymph node metastasis(50.00%) was much lower than that with lymph node metastasis(82.98%), both of them have statistical significance(P<0.05).3. Significant difference of metastin and MMP-9 expression in gastric mucosa tissues could be seen(P<0.01). It was a negative correlation between them signifi- cantly(r=-0.365 ).Conclusion:1. The expression of KiSS-1 peptide metastin in gastric carcinoma tissues was significantly lower than that in gastric mucosa tissues from radical surgical margin and gastric mucosa tissues from surgical margin in patients with benign diseases, and was decreasing with the advancement of clinical stage. Expression of metastin in patients with gastric carcinoma associated with lymph node metastasis was significantly lower than that without lymph node metastasis. 2. The expression of MMP-9 in gastric carcinoma tissues was significantly higher than that in gastric mucosa tissues from radical surgical margin and gastric mucosa tissues from surgical margin in patients with benign diseases, and was increasing with the advancement of clinical stage. Expression of MMP-9 in patients with gastric carcinoma associated with lymph node metastasis was significantly higher than that without lymph node metastasis.3. No correlation was found between the expression of metastin and MMP-9 in gastric carcinoma tissues with its pathological types.4. There was a negative correlation between the expression of metastin and MMP-9 in gastric carcinoma significantly.
