The paper aimed at revealing the active substance agreeing with primary dysmenorrheal in Commiphora myrrha Eng1.In the first chapter,a detailed review was given to the latest development about formation mechanism,the correlation between effectiveness and active substance of resins.Besides,chemical constituents from resin of Commiphora species and their biological activities,and the latest development about primary dysmenorrheal was also summarized.In the second chapter,the active substance on treating primary dysmenorrheal was screened out.The pharmacological models,including primary dysmenorrheal mice model,pathological section,analgesic experiment,anti-inflammatory experiment,platelet aggregation test,and so on, were adopted to evaluating the effects of myrrha and its different separated fractions.The results were listed as follow.1.The results on primary dysmenorrheal mice model stated that Extraction G and F can reduce the writhing times,increase nitric oxide concentration and reduce calcium ion concentra -tion in uterine tissue of mice much significantly.The volatile oil of myrrha can reduce the writh -ing times significantly.2.Investigation on pathological section of primary dysmenorrheal mice show that Extrac -tion G can improve the uterine pathological significantly.The edema and increased inflamma -tory cells were not observed in pathological section of mice in Extraction G group.3.The results of analgesic experiment stated that Extraction G,F and the volatile oil showed significant analgesic activity.However,Extraction G,F and the volatile oil showed no activity on hot plate test.4.The results on anti-inflammatory experiment showed that Extraction G.F and the volatile oil can inhibit edema by subcutaneous injection of formaldehyde and reduce the content of PGE2 in inflammation tissue.5.The effects on platelet aggregation induced by ADP stated that Extraction A,F,G.H showed inhibitory activity on platelet aggregation induced by ADP(IC50:56.80 mg/mL,28.68 mg/mL,12.40 mg/mL,25.00 mg/mL).The volatile oil showed no inhibitory activity on platelet aggregation.6.The results on cell proliferation stated that Extraction B,F show inhibitory activity on shikawa,A2780,SiHa,SK-OV-3 cell.The results of inhibitory activity of Extraction B: shikawa(IC50:26.634ug/ml),A2780(IC50:26.908ug/ml),SiHa(IC50:32.204ug/ml),SK-OV-3 (IC50:38.716ug/ml).The results of inhibitory activity of Extraction F:A2780(IC50:15.809ug/ml), shikawa(IC50:20.733ug/ml),SK-OV-3(IC50:22.076ug/ml),SiHa(IC50:57.830ug/ml).Inhibitory activity of myrrha on cell proliferation and sesquiterpenes is closely related.In the third chapter,the chemical components of myrrha were studied.Two compounds were isolated and identified from Extraction G as 2-methoxy-8,12- epoxygermacra-1(10),7,11-trien -6-one(1),2-Methoxy-5-acetoxy-fruranogermacr-1(10)-en-6-one(2).Four compounds were isolated and identified from Extraction H as Mansumbinone(3),myrrhone(4),behenic acid(5),β-sitosterol(6).In the four chapter,the method of UPLC-MS/MS and GC-MS was adopted to screen and analyze the potential active components.1.The results of analysis by GC-MS showed that there were 41 components in about 57.22%of the total essential oil.The main components of essential oil were sesquiterpenes.The analgesic activity and sesquiterpenes is closely related.2.The results of analysis by UPLC-MS/MS showed that The main components of Extrac -tion G and F were sesquiterpenes,diterpenes.The terpenoid maybe the active substance on treating primary dysmenorrheal.
