The Effect Of Zhuang’s Herbal Moxibustion On The Expression Of NF-kB And TNF-α In Rats With Primary Dysmenorrheal

Objective: To observe the effect of Zhuang’s herbal moxibustion on Ear engorgement、Threshold of pain and the expression of the NF-kB 、TNF-α and related inflammatory factors in Model Rats with primary dysmenorrheal,and investigate the possible mechanism of Zhuang’s herbal moxibustion in the treatment of the dysmenorrheal model.Methods:60female SD rats with sexual maturity(10-12 weeks)were selected.They were randomly divided into normal group(Group A),Model group(Group B),moxibustion group(Group C),Zhuang’s herbal moxibustion group(Group D),guizhifuling group(Group E),acupuncture group(Group F).The model was made by cold wather and injection of estradiol,adrenaline and oxytocin in the abdomen.After making the model,the pain threshold and ear engorgement of each group were detected after the intervention.At the same time,the amount of IL-2 and VEGF in serum and uterus were detected by ELISA.The expression of NF-kB and TNF-αin uterine were detected by immunohistochemistry.Results:C group,D group,E group and F group could significantly reduce the concentration of VEGF 、 NF-kB and TNF-α in rats with dysmenorrheaand increase the expression of IL-2(compared to dysmenorrhea model group P < 0.05).After treatment,the tolerance to thermal pain was higher in the treatment groups than in the model group.Compared with the other three treatment groups the D group’s concentration of VEGF in serum and tissue of rats was lower than group C(P<0.05)The expression of NF-kB in D group was higher than that in E group and F group was lower(P<0.05);the expression of TNF-α in group D was lower than that in group C(P<0.05),and there was no significant difference between the other indexes(P > 0.05).Conclusions: the occurrence of dysmenorrhea may be related to the abnormal expression of the NF-KB pathway of the tissue,the mechanism of Zhuang’s herbal moxibustion in the treatment of dysmenorrhea may be related to inhibiting the activation of NF-kB pathway,reducing the release of inflammatory factors and relieving the uterine spasm;Reducing blood vessels leakage and blood clotsand;Reducing neuropathic pain and elevate pain threshold.The overall performance in rats after treatment of pain threshold changes better than the model group.
