Objective: To investigate the level of B7-H1 expression in the human gastric cancer, and its correlationship with age, sex, location and the clinical pathological indicators such as the size, location, depth of invasion, differentiation and distant metastasis of tumor.Methods: A total of 45 clinical samples were obtained for investigation. In 15 of them samples were taken from the gastric carcinoma and its adjacent normal tissue respectively for each one. Western Blot was used to investigate the expression of B7-H1 in gastric carcinoma and its adjacent normal tissue, in which the expression of B7 -H1 molecule on the surface of macrophages was measured using flow cytometry as well. The other 30 samples were only taken from gastric cancer and the expression of B7-H1 molecule on the macrophages were studied by flow cytometry. Then, the results obtained were used to analyze the correlationship between the expression of B7-H1 and clinical indicators in association with clinical pathological data.Results: 1.The data of Western Blot indicated that B7-H1 molecule can be detected in all of the patients with gastric carcinoma and its expression level is significantly higher in gastric carcinoma than in its adjacent normal tissue. 2. B7-H1 was found to express on the surface of macrophages from both gastric cancer and adjacent normal tissue by flow cytometry and its expression were significantly higher in carcinoma compared to its adjacent normal tissue with percentages of 20.71±3.69% and 6.83±1.80% repectively(P <0.01)。3 From the flow test results it can been seen that the high level of expression of B7-H1 was significantly correlated with the existence of distant metastasis in patients (P <0.05) instead of gender and age of patients, location, size, depth of invasion and differentiation of tumor, (P> 0.05).Conclusion: 1.B7-H1 could be used as an independent indicator of clinical prognosis and by analyzing B7-H1 the effective treatment can be delivered to patients with high risk and poor prognosis as soon as possible to inhibit the disease from deteriorating. 2. The antagonists of B7-H1 can be applied to block the transmission of inhibitory signals in B7-H1/PD-1 pathway, as might become a new target for cancer immunotherapy.
