Objectives:To explore the expression of COX-2 and Ki-67 in Hepatobiliary Calculus with Cholangiocarcinoma(HCWC) and its clinical significance.Relationship between the expression of COX-2,Ki-67 and the onset and progress of HCWC was investigated to lay an experimental base for the diagnosis and treatment of HCWC.Methods:Eighteen cases of normal tissue of bile duct from operations of hemangiomas of liver or liver trauma as the control(group A),fifty five cases of the tissue of bile duct surrounding calculus of intra-hepatic(group B) and forty four cases of tumor tissue of HCWC(group C) were randomly sampled and collected in Hepatobiliary Surgery in Hunan Provincial People's hospital between January 2000 to December 2009; Immunohistochemistry was employed to detect and statistically analyze the expression of COX-2 and Ki-67 in each group.Results:1. In group A,B,C,the positive rate of the expression of COX-2 was 5.56%,32.72% and 70.45%,respectively.The positive rate of the expression of COX-2 in the carcinoma tissue of HCWC was significantly higher compared with that in the control group(P<0.05);the positive rate of the expression of COX-2 in poorly differentiated HCWC cases was higher compared with that in the highly differentiated cases(P<0.05);the positive rate of the expression of COX-2 in extra-hepatic or lymphatic metastasis HCWC cases was higher compared with that without metastasis cases(P<0.05).The expression of COX-2 in the carcinoma tissue of HCWC was not relative to the gender,age,course,tumor size and tumor pathological types(P>0.05).2.In group A,B,C the positive rate of the expression of Ki-67 was 0,27.27% and 79.55%,respectively.The positive rate of the expression of Ki-67 in the carcinoma tissue of HCWC was significantly lower compared with that in the control group(P<0.05);the positive rate of the expression of Ki-67 in poorly differentiated HCWC cases was lower compared with that in the highly differentiated cases(P<0.05);the positive rate of the expression of Ki-67 in extra-hepatic or lymphatic metastasis HCWC cases was lower compared with that without metastasis cases(P<0.05).The expression of Ki-67 in the carcinoma tissue of HCWC was not relative to the gender,age,course,tumor size and tumor pathological types(P>0.05).3.The expression of COX-2 was followed by a high expression of Ki-67 in the tissues of HCWC. Spearman-related analysis show a strong positive correlation between the expression of COX-2 and that of Ki-67 in HCWC(r=0.413,P=0.005).Conclusions:1.High expression of COX-2 is related to the occurrence and progress of HCWC; 2. High expression of Ki-67 is related to the occurrence and progress of HCWC;3.There is a positive correlation between the expression of COX-2 and that of Ki-67 in HCWC.
