Objective To explore the expression of cyclooxygenase-2 (COX-2), phosphatase and tensin homolog deleted on chromosome ten (PTEN) in Hepatobiliary Calculus with Cholangiocarcinoma (HCWC) and its clinical significance. Relationship between the expression of COX-2, PTEN and the onset and progress of HCWC was investigated to lay an experimental base for the prevention and treatment of HCWC.Materials and Methods Thirty seven cases of tumor tissue of HCWC (group C), thirty cases of the tissue of bile duct surrounding calculus of intra-hepatic (group B), and ten cases of normal tissue of bile duct from operations of hemangiomas of liver or liver trauma as the control (group A) were randomly sampled and collected; two-step immunohistochemistry (SP method) was employed to detect and statistically analyze the expression of COX-2 and PTEN in each group.Results1. In group A,B,C ,the positive rate of the expression of COX-2 was 10% ,33.3%, and 70.3% , respectively. The positive rate of the expression of COX-2 in the carcinoma tissue of HCWC was significantly higher compared with that in the control group(P<0.01); the positive rate of the expression of COX-2 in poorly differentiated HCWC cases was s??ficantly higher compared with that in the highly differentiated cases (P<0.05); the positive rate of the expression of COX-2 in extra-hepatic or lymphatic metastasis HCWC cases was significantly higher compared with that without metastasis cases(P<0.05).. The expression of COX-2 in the carcinoma tissue of HCWC was not relative to the gender, age, course(P>0.05). 2. In group A,B,C the positive rate of the expression of PTEN was 90.0%, 80.0%, and 35.0%, respectively. The positive rate of the expression of PTEN in the carcinoma tissue of HCWC was significantly lower compared with that in the control group (P<0.01); the positive rate of the expression of PTEN in poorly differentiated HCWC cases was significantly lower compared with that in the highly differentiated cases (P<0.05);the positive rate of the expression of PTEN in extra-hepatic or lymphatic metastasis HCWC cases was significantly lower compared with that without metastasis cases(P<0.05). The expression of PTEN in the carcinoma tissue of HCWC was not relative to the gender, age, and course (P>0.05).3. The expression of COX-2 was followed by a low expression of PTEN in the tissues of HCWC. Kendall-related analysis show a strong negative correlation between the expression of COX-2 and that of PTEN in HCWC (r=-0.323, P <0.05).Conclusion1. High expression of COX-2 may be related to the occurrence of HCWC;2. There may be a negative correlation between the expression of COX-2 and that of PTEN in HCWC;3. High expression of COX-2 and low expression of PTEN may be prompted to HCWC in extra-`攧tic or lymphatic metastasis.
