Objective:In order to study the chronic obstructive pulmonary disease occurrence and development of the pathological process furtherly and provide new ideas for clinical prevention and treatment. We investigate the expression and correlationof Hypoxia inducible factor-1α(HIF-1α) and Vascular Endothelial Growth Factor (VEGF) in rats model with chronic obstructive pulmonary disease (COPD) and the chronic bronchitis.Methods:Healthy male Wistar rats were randomly divided into three groups:normal control group,chronic bronchitis group, and COPD group. Normal group, under normal circumstances and normal feeding for 14 weeks Cigarette somoking was used to build the mode of the chronic bronchitis and COPD model, chronic bronchitis group were smoked 7weeks,COPD group of 14 weeks. After the end of times smoked, rats in each group respectively Weighing and take their right middle lobe.The cases were stained by HE staining and diagnosed by more than two senior pathological professors. we detected the expression of HIF-1α, VEGF by immunohistochemical staining.All dates were analysis by software SPSS 13.0.Results:Rat COPD and chronic bronchitis were established successfully by exposure to cigarette smoke,which were confirmed by the indices of pulmonary emphysema. COPD rats are obviously lighter than the normal rats.The HIF-1αand VEGF was positively expressed in bronchioles and arterioles of COPD and chronic bronchitis rat model compared with control group(P<0.05). The expression of HIF-1αand VEGF was no significant difference between the chronic bronchitis and COPD (P>0.05). There was a positive correlation between the HIF-1αand VEGF in three groups (P<0.01).Conclusion:1)Our study showed that rat chronic bronchitis and COPD model can be established successfully by exposure to cigarette smoke daily.(2) COPD rats are obviously lighter than the normal rats,showing the COPD rats may undernourished.(3) There is a close correlation between HIF-1αand VEGFand the chronic obstructive pulmonary disease. (4) There was a positive correlation between the HIF-1αand VEGF in three groups,showing HIF-1αand VEGF may anticipate the airway inflammation and remodeling in COPD togethert.
