ObjectiveDo the experiment on resisting Non-alcoholic fatty liver to appraise the hepatoprotective effectof Chinese herbal compound XIAOZHIGTANG(XZT),and to study the mechanism of theeffect.Animal model,whose livers were damaged by D-GaIN,was used to study the Anti-lipid peroxidation effect of XIAOZHIGTANG.Experimental contents1.Experimental study on the anti-rat NASH effect of XIAOZHITANG.2.Protective effects of XZT on the model of Fatty Liver induced by Tetracycline in Mouse.3.Protective effects of XZT on the model of Fatty Liver induced by DL-ethionin in Mouse.4.Protective effects of XZT on the model of the liver injury induced by D-GalN in Mouse.Methods1.Fouty SD rats were randomly divided into 4 groups,normal group(groupA),model group(groupB),group of XZT(groupC)and group of fenofibrate(groupD).The rats in normal group were fed with normal diet,and the rats in other groups were fed with fat emulsion.After three weeks,fatty liver model was established. Then,give groupC and groupD the treatment,and at the same time,continue to feed the three groups with fat emulsion.The treatment will be continued for four weeks.And at the twelve hours after the last administration of the drugs,rats in all groups would be killed.Blood samples for detecting ALT,AST activity and TG,TC contents were collected.Histopathological examintion was also performed for liver tissue.2.Sixty KunMing mice were randomly divided into 6 groups,nomal group(groupA), model group(groupB),high dosage group of XZT(groupC),middle dosage group of XZT(groupD),small dosage group of XZT(groupE)and fenofibrate group (groupF).Except normal group(groupA),from the first day,the other groups was administered with drugs,once a day.Then,on the eighth day,except normal group(groupA),mice in other groups were injected tetracycline to establish the fatty liver model,and the injection would be continued for four days.One hour after the last administration of the drugs,liver tissues were collected for measuring the level of the hepatic TG and hepatic TC,and for calculating the liver quotiety.Then take a histopathologic examination of the liver tissues.3.Sixty KunMing mice were randomly divided into 6 groups,nomal group(groupA), model group(groupB),high dosage group of XZT(groupC),middle dosage group of XZT(groupD),small dosage group of XZT(groupE)and fenofibrate group (groupF).Except normal group(groupA),from the first day,the other groups was administered with drugs,one time a day,it will be continued for four days.six hours after the last administration of the drugs,except groupA,mice in other groups were administratered with DL-ethionin 250mg/kg.After twenty four hours,Blood samples for detecting TG and TC contents were collected,and liver tissues were collected for measuring the level of the hepatic TG and hepatic TC,and for calculating the liver quotiety.Then take a histopathologic examination of the liver tissues.4.Sixty NIH mice were randomly divided into 6 grOups,normal group(groupA), model group(groupB),high dosage group of XZT(groupC),middle dosage group of XZT(groupD),small dosage group of XZT(groupE)and bifendate group (groupF).Mice in groupC,groupD,groupE and groupF were administered with drugs by gastric perfusion for 7 days once daily consecutively.One hour after the last administration of the drugs,excepts those in group A,mice in other groups were injected with D-GalN 800mg/kg into its abdominal cavity.Blood samples for detecting ALT,AST,superoxide dis-mutase(SOD)activity and malondiadehyde(MDA)concentration were collected at the 16 hours after D-GaIN injected.Histopathological examintion was also performed for liver tissue.Results1.The XZT could significantly lower serum ALT;AST activity,and lower serum TG,TC contents.In the liver tissue,XZT could lower the TG,TC centents as well.And it could markedly ameliorate the liver damage(P<0.05).2.In the experiment of Protective effects of XZT on the model of Fatty Liver induced by Tetracycline in Mouse,the high dosage XZT and the middle dosage XZT could lower the TG and TC contents in the liver tissue,lower the liver quotiety and significantly improve the histopathologic change of the liver tissues(P<0.05).3.The high dosage XZT and the middle dosage XZT could significantly depress the content level of TG and TC in liver tissue which was damaged by DL-ethionin.And they could also lower the liver quotiety(P<0.05),and hepatic histopathological change were alleviated in both XZT with high dosage and middle dosage.But to the serum TG and TC contents,XZT have no significant effect(P>0.05).4.The high dosage XZT and The middle dosage XZT significantly depressed serum ALT,AST and MDA in the liver tissue,which was damdaged by D-GalN.And they also enhanced SOD in the liver tissue and markedly ameliorated the liver damage(P<0.05).ConclusionChinese herbal compound XIAOZHITANG can protect liver against NASH which could induced by different factors.Maybe,the mechanism is the compound can enhance the elimination of OFR.
