| Objective Invation and metastasis is the primary biologic character of process in tumor,is also the important reason of relapse and metastasis after operation.In the past years,many biologic studys analyze that gene and molecule events in cancer can prove the commencement,development and control of cancer is related with gene.The extensive develop of the research about gene accelerating or controling tumour has bring on the new gene reasearch that can control growth,spread and metastasis of cancer.Gastric cancer is still a malignant tumor which incidence rate and mortality rate accounts for the first. Seventeen thousand people were died of gastric cancer every year.At present 90% of inpatient cases were advance period cancer.The five year survival rate loiters at 30%-40%.Even if radical operation was done,metastasis is the main problem to influence the prognosis.It is the key point to improve curative effectiveness which study metastasis regulation,build early age diagnosis of sub-clinical metastasis or forecast guide line and therapy targets.CD82 KISS-1 is a metastasis suppressor gene,they have been found in the recent years.Their activeness is about with metastasis and incoachment of many malignancy. But it was not widely studied in gastric cancer..The research will discuss the ralation of CD82 KISS-1 gene and the clinic character and relapse and metastasis in gastric cancer through detecting the expression in gastric cancer.Method:50 patients' post opertation paraffin specimens of heze municipal hospitai in 2002 were recruited by chance.24 males,26 females,from 20 to 69 years,the middle year is 40, It can be classified papilloadenocarcinoma cannular adeno carcinoma low-grade differentiation adenocarcinoma mucilage adenocarcinoma signet-ring cell carcinoma by histology model.and papilloadenocarcinoma cannular adenocarcinoma mucilage adenocarcinoma were classified high, middling-grade differentiation group, signet-ring cell carcinoma low-grade differentiation adenocarcinoma were low-grade differentiation group.According to the new TNM stage by international union control cancer(IUCC),Ⅰa 1,Ⅰb 2,Ⅱ5,Ⅲa 12,Ⅲb 16,Ⅳ14,36 lymph node metastasis,6 viscera metastasis,13 in gastic antrum,26 in cardia,11 in gastic curve,the diameter is 2.0-7.0 centimetre,no chemotherapy were executed before operation, all were visited at any moment in 3-5 years.all samples were analyzed by immunohistochemical method,the expression of CD82 KISS-1 gene was contrasted with natural gastic tissue. Statistics analyse was done basis on clinic data and visit data.Result:(1)45 out of 50 cases with normal gastric mucosas were CD82 positive, the positive rate was 90%.Amang 50 cases with gastric cancer,the positive cases of CD82 were 6,the weak positive cases were 6,the negative cases were 38,the positive rate was 24%,the difference is significant between them.(P>0.05) 43 out of 50 cases with normal gastric mucosas were KISS-1 positive,the positive rate was 94 %.The positive cases of KISS-1 amang 50 cases with gastric cancer were 6,the weak positive cases were 10,the negative cases were 34,the positive rate was 32%. the difference is significant between them.(P>0.05)(2)The expression of CD82 in cancers with low-grade differentiation lymph node metastasis viscera metastasis was evidently fallen(P<0.05),the positive expression rate of CD82 in the patients who survived more than 5 years was higher than that of in the patients \ who survived less than 5 years.(P<0.05),the five year survival rates of the patients with positive weak positive negetive expression of CD82 decreased in turn 83%50%20%.,the expression of KISS-1 in cancers with low-grade differentiation lymph node metastasis viscera metastasis was evidently fallen(P<0.05),the positive expression rate of KISS-1 in the patients who survived more than 5 years was higher than that of in the patients who survived less than 5 years(P<0.05),the five year survival rates of the patients with positive weak positive negetive expression of KISS-1 decreased in turn 83%70%12%.Conclusion:The expression of CD82 KISS-1 in gastric cancer is lower than in normal gastric mucosas,it indicated the occurrence of gastric cancer is related with the absence of CD82 KISS-1. The expression of CD82 KISS-lin gastric cancer was not correlative with age borrmann type tumor size site pathological type,but had statistics meaning with differentiation degree invasive depth lymph node metastasis. The expression of CD82 KISS-1 in gastric cancer was reduced while differentiation degree degraded,reduced while invasive depth deepened,the positive rate in gastric cancer with lymph node metastasis was lower distinctly than in gastric cancer without lymph node metastasis, the five year survival rates in positive expression of gastric cancer was higher than in nagative expression.it cued their expression paticipated the control of gastric cancer metastasis probably,suppressed the metastasis effectively.the gene CD82 KISS-1 became the new biological marker which forecast the malignant potential of gastric cancer,the new pathway of genic treatment will be come true by restoring and improving genic expression level in our body. |
