The Experiment About The Effect Of The Acup-uncture Plus Heat On The Nerve Growth Factor In The Chrpmoc Alcoholic Peripheral Neuropathy Rats

Objective:We use a free-drink model on peripheral neuropathy of chronic alcoholism and discuss the possible mechanism of effects acupuncture plus heat on CAPN through experimental research, which can provide the new theoretical basis for clinical expansion of the acupuncture treatment of chronic alcoholic peripheral neuropathy.Method: Seventy Wistar rats are divided into two groups randomly: control group and experiment group. The control group drink water. The alcoholic concentration of experiment group were increased from 6%, 9%, 12%to20% every five days, and the rat were breed with 20% alcoholic concentration until the end. The rat were examined with pressure pain threshold,EMG and pathology by the end of two, three, and four month. We divided the models into 4 groups randomly.They are model group, methycobal -control group,and acupuncture plus heat group. We observed the effect of acupuncture plus heat treatment on weight,pressure pain threshold,sciatic nerve conduction velocity,pathomorphology of sciatic nerve and dorsal rootganglia , NGF level in sciatic nerve and dorsal rootganglia on model rats by the technic methods of electrophysiology,pathology, immuno-histochemistry,elisa and molecular biology.Results: 1,The increase of body weight in model group was obviously slower than that in control group.2,The pressure pain threshold in model group was significantly highter than that in control group after the end of three month (P<0.05).3,Electrophysiologically the model group rat showed slow motor conduction velocity (MCV) by the end of four month, compaired with control group(P<0.01).4,HE stain of sciatic nerve of the rat at fouth month end in light microscope showed axonal degeneration accompany with secondary segmental demyelination, but the structure of ganglia showed non apparent abnormality.5,Acupuncture plus heat treatment can improve the pressure pain threshold and the nerve conductive velocity,reduce the changes of pathological morphology and ultrastructure in sciatic nerve and increase the expression of NGF in sciatic nerve and DRG. Compared with methycobal-control group ,the difference were significant ( P<0.05) .Conclusions: 1. Acupuncture plus heats may reduce the CAPN rat 's pressure threshold of feeling.2. Acupuncture plus heat treatments has the function of improvement the CAPN rat sciatic nerve conduction speed.3.The acupuncture plus heat treatment may improve the CAPN rat sciatic nerve textile fiber to escape the myelinic membrane and the axis cylinder withers, reduces the nerve the pathology damage, has the protective function to the nerve.4. Acupuncture plus heats may promote CAPN rat sciatic nerve and in the back root ganglion the nerve growth factor expression.
