Influence Of "Shuanggu Yitong" Moxibustion On Neurological Function In Rats With Type 2 Diabetic Peripheral Neuropathy

AimsStudy on the influence of "Shuanggu Yitong" moxibution on blood sugar, sciatic nerve conduction velocity (NCV) and nerve growth factor (NGF) protain expression in the rat model of diabetic peripheral neuropathy (DPN), Discuss on the function mechanism of moxibustion on preventing and curing DPN basing on the neunotrophins're-restoration to injured nerve.MethodsUse 50 200±20g Wistar rat, select 10 out of them by random as normal control group (Group A). The other 40 rats are established as diabetes mellitus (DM) rat models by peritoneal injection with streptozotocin (STZ). Get the successful rats which the fasting blood glucose lever above 16.7mmol/L, randomly divide them into 4 groups:DM model group (10 rats, Group B); DM curing group (10 rats, Group C); DPN model group (10 rats, Group D); DPN treatment group (10 rats, Group E). According to "Shuanggu Yitong" treatment principle, select the rats from Group C to moxibustion on "Guanyuan", "Zusanli" and "Yishu" with 15 minutes every day and last for 4 weeks. Contemporary as the treatment of Group C, after 4-week's normal feed, there's a significant decrease in sensory nerve conduction velocity (SNCV) and motor nerve conduction velocity (MNCV) of rats from Group D and E, and those rats turn into DPN rat models. According to "Shuanggu Yitong" treatment principle, select the rats from Group E to moxibustion on "Guanyuan", "Zusanli" and "Yishu" with 15 minutes every day and last for 4 weeks. At the second day of the last moxibustion, cut the rats'tails and measure their fasting blood glucose, and then measure the NCV of rats from each Group with electrophysiology method; then anaesthetize each group's rats, quickly separate the sciatic nerve of them, and immersed the hippocampus into 4% paraformaldehyde in standby, measure the NGF protain expression of rats from each Group with immunohistochemical SABC method.Results1.Compared with Group A, there's an increase in blood sugar level of group B,C,D,E, with a significant discrepancy on statistical analysis (P<0.05); After "Shuanggu Yitong" moxibution's curing, Compared with Group B, there is a significantiy decrease in blood sugar level of Group C (P<0.05); Compared with Group D, there is a decrease in blood sugar level of Group E (P<0.05);2.Compared with Group A, there's a decrease in SNCV and MNCV of group B,C,D,E (P<0.05); Compared with Group B, there is an increase in NCV of Group C (P<0.05); Compared with Group D, there is an increase in NCV of Group E (P<0.05);3.Compared with Group A, there's a decrease in NGF positive expression of group B and D model rats with a significant discrepancy on statistical analysis (P<0.05); Compared with Group B, there is an increase in NGF positive expression of Group C (P<0.05); Compared with Group D, there is an increase in NGF positive expression of Group E (P<0.05).Conclusions1. "Shuanggu Yitong" moxibustion can effectively adjust the blood glucose level of DM and DPN rats, reduce the dysfunction of peripheral nerve from the higher blood glucose.2. "Shuanggu Yitong" moxibustion can speed up the DM and DPN rats' SNCV,MNCV, also can improve peripheral nerve structure and rehabilitate the nerve function.3."Shuanggu Yitong" moxibustion can raise NGF positive expression of each group, take the role of repairing damaged nerve.
