Effects Of San Wubai Powder On Proliferation And Apoptosis Of Human Gastric Cancer Cell Line Sgc-7901

Objective:In vitro experiments in rats to explore SanWuBai Powder containing serum on human gastric cancer SGC-7901 cell proliferation, apoptosis inhibition and its mechanisms, and thus to explore the treatment for gastric cancer based on cellular and molecular biology.Methods:1. Animal experiment:Select 20 SPF-class male rats, all the rats were randomly divided into four groups,five rats of each group:the low-dose SP group(0.035g/kg.d),the middle-dose SP.group(0.07 g/kg.d),the high-dose SP group(0.14 g/kg.d) and the saline group. Application of acridine orange, EB staining measured in different concentrations of rat serum containing SanWuBai Power of human gastric cancer SGC-7901 cells, the role of apoptosis 48h after the impact.Results:(1) MTT assay results showed that:low, middle and high doses of SanWuBai Power of rat serum containing a proliferation of human gastric cancer SGC-7901 cells were inhibited. Different concentration groups SanWuBai Power-containing serum-treated cells 12-72h after the OD ratio of each group were decreased in varying degrees. With SanWuBai Power and the dose of drug-containing serum increases, the role of the extension of the time, OD ratio gradually decreased, the survival rate gradually decreased, that is, SanWuBai Power-containing serum on the proliferation of SGC-7901 cells in a dose-inhibition concentration and time-dependent, and the inhibitory rate of high-dose SanWuBai Power the role of drug-containing serum 48h highest up to 69.2%.(2)As amended by three-dose group SanWuBai Power serum-containing cells after 48h treatment, low, middle and high doses SanWuBai Power-containing serum 48h role in SGC-7901 cells, the apoptosis percentage of each group followed by 25%,32%,54%,and with the concentration increased, the extension of the time increased. SanWuBai Power can restrain the apoptosis of SGC-7901 cells. High concentration of SanWuBai Power (20% High concentration) have the time-effect to SGC-7901 cells.Conclusions:1. SanWuBai Power in a certain dose range, can inhibit the proliferation of SGC-7901 cells, and showed a certain dose, concentration and time-dependent.2. SanWuBai Power can induce apoptosis, the effect was a definite dose and time-dependent.3 This study SanWuBai Power in the application of anti-gastric cancer provides a theoretical basis.
