Objective Detection of miR-593-5p expression in gastric cancer tissues, analyzed with the tumor size, infiltration, lymph node metastasis, tumor staging and other relations, guide up regulation of miR-593-5p expression by lentivirus mediated, observe the effect of overexpression of miR-593-5p on proliferation and apoptosis of human gastric cancer SGC-7901 cells, To investigate the biological role of miR-593-5p in the development and progression of gastric played.Methods The collection of specimens from patients with gastric carcinoma in 58 cases, at the same time take cancer and corresponding cancer adjacent non tumor tissue. The expression level of miR-593-5p in each tissue fluorescence quantitative PCR detection, the expression of miR-593-5p ingastric cancer tissues and paracancerous tissues. Analysis of miR-593-5p expression with age,gender, tumor size, infiltration, lymph node metastasis, tumor staging and other relationship. The use of recombinant lentiviral particles carrying miR-593-5p (LV-miR-593-5p-G V) overexpression, plasmid lentivirus (LV-GV) infection of human gastric cancer SGC-7901 cells, respectively, as the experimental group (LV-miR-593-5p-GV), negative control group (LV-GV), human gastric carcinoma cell line SGC-7901 transfected processing as the blank group, the above three groups of cells in the conventional culture, CCK-8 method and flow cytometry to detect the effect of overexpression of miR-593-5p on proliferation andapoptosis of human gastric cancer cells SGC-7901.Results Volume was significantly lower than the adjacent cancer tissue miR-593-5p expression in gastric cancer tissues, the difference was significant (P<0.05). Expression of miR-593-5p in gastric cancer with tumor stage, tumor size, infiltration, patient age, gender, lymph node metastasis and other clinical factors showed no significant correlation. Compared with the negative group and the blank group, the experimental group of human gastric cancer SGC-7901 cell proliferation ability reduced, apoptosis rate increased, the difference was statistically significant (P<0.05), while the negative control group and blank group, no significant difference (P>0.05).Conclusions Expression of miR-593-5p in gastric cancer tissue is lower than the normal tissues adjacent to carcinoma, The expression of gastric cancer associated with tumor staging. miR-593-5p overexpression can inhibit the proliferation of human gastric cancer SGC-7901 cells, suggesting that miR-593-5p and related to the occurrence and development of gastric cancer.
