Objective:To investigate the effects of Traditional Chinese Medicine Qingrequshi Decoction on TNF-αand insulin resistance in patients and model rats with type 2 diabetes. Methods:1. Clinical Study:80 patients of type 2 diabetes with damp-heat syndrome were randomly assigned to two groups, therapentic group(50 cases) treated with Traditional Chinese Medicine Qingrequshi Decoction, control group(30 cases) treated with metformin hydrochloride, The course was 2 months. TNF-α, BMI, blood lipid, blood rheology, HbA1C, fasting plasma glucose, postprandial plasma glucose and fasting insulin were observed before and after treatment. Insulin resistance was evaluated by IAI and HOMA-IR. Results After 2 months treatment of Traditional Chinese Medicine Qingrequshi Decoction, the TNF-αwas reduced significantly, the clinical symptoms was improved, the blood fibrinogen, HbA1C, fasting plasma glucose, fasting insulin and the blood lipid was reduced significantly, IAI and HOMA-IR was improved. The difference was significant compared to the control group.2. Experimental Study:90 male wister rats was selected.Model rats of insulin resistance were established by feeding with high fat forage combined with intraperitoneal injection of small dose of streptozotocin.50 model rats were randomly assigned to 5 groups, high dose group, morderate dose group and low dose group were treated with Traditional Chinese Medicine Qingrequshi Decoction, control group was treated with metformin,model rats group and normal group were treated with distilled water.The course was 2 months. The TNF-α, blood lipid, fasting plasma glucose and fasting insulin were observed before and after treatment. Insulin resistance was evaluated by IAI and HOMA-IR. Results After 2 months treatment of Traditional Chinese Medicine Qingrequshi Decoction, TNF-α, blood lipid,fasting plasma glucose and fasting insulin was reduced obviously, IAI and HOMA-IR was improved obviously. The difference was significant compared to control group. And the high dose group improved significantly. Conclusion Traditional Chinese Medicine Qingrequshi Decoction could reduce the blood TNF-α, and improve the metabolic disorder and ameliorate the degree of insulin resistence in patients of type 2 diabetes with damp-heat syndrome and model rats with type 2 diabetes.
