Study The Expression And Significance Of Brms1, Gelsolin And Upa In Gastric Carcinoma | | Posted on:2011-02-11 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y L Zhang | Full Text:PDF | | GTID:2194330332470375 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | ObjectiveBy investigating the expression of BRMS1, Gelsolin and uPA in gastric carcinoma and adjacent normal tissue,to explore the relationship between the expression pattern of protein and the biological behavior in gastric carcinoma.This study also analysis the correlation of the expression of these three kinds of protein in order to explore the molecular mechanisms biology for metastasis of gastric carcinoma.This study would look for the new markers for evaluating biological characters and prognostic evaluation of gastric carcinoma.MethodsThe expression of BRMS1,Gelsolin and uPA were detected by immunohistochemical method among the 48 cases of paraffin-embeded gastric carcinoma and the adjacent mucosa.Then we analyzed their relationships and correlations with patient's age,sex,degree of differentiation,histological type,lymph node metastasis and other clinicopathologic findings.The results were treated by x2 test and Spearman Correlation analysis.Results1. The positive BRMS1 rate of expression in 48 cases with gastric carcinoma is 47.9%(23/48),while that of the adjacent tissue is 95.8%(46/48).The positive rate of BRMS1 was higher in adjacent mucosa than that in gastric carcinoma.There is significant difference between the two groups (P<0.01).There is significant correlation between the expressions of BRMS1 with the lymph node metastasis and TNM stage (P<0.05).However,there is no relationship between the expression of BRMS1 with patient's age, sex, location of tumour, size,degree of differentiation and depth of infiltration (P>0.05)2. The expression of Gelsolin in 48 cases with gastric carcinoma is 31.3% (15/48),while that of the adjacent tissue is 83.3%(40/48).The positive rate of Gelsolin was higher in adjacent mucosa than that in gastric carcinoma.There is significant difference between the two groups (P<0.01).There is significant correlation between the expressions of Gelsolin with the lymph node metastasis and depth of infiltration (P<0.05).However,there is no relationship between the expression of Gelsolin with patient's age,sex,location of tumour, size,degree of differentiation and TNM stage (P>0.05)3. The positive uPA rate of expression in 48 cases with gastric carcinoma is 70.8%(34/48),while that of the adjacent tissue is 10.4%(5/48).The positive rate of uPA was higher in gastric carcinoma than that in adjacent mucosa.There is obvious difference between the two groups (P<0.01).There is significant correlation between the expressions of uPA with the lymph node metastasis,depth of infiltration,degree of differentiation and TNM stage (P<0.05).However,there is no relationship between the expression of Gelsolin with patient's age, sex, location of tumour and size (P>0.05)Conclusions1. BRMS1 protein expression in gastric cancer tissues was significantly reduced than that in adjacent tissue.The expression of BRMS1 protein significantly correlated with the lymph node metastasis and the stage of TNM.This suggested that BRMS1 protein may be used as the biological assessment of metastasis of gastric cancer indexes,the reduction in its expression can promote metastasis of gastric cancer.2. Gelsolin protein expression in gastric cancer tissues was significantly lower than that in adjacent normal tissue.The expression of Gelsolin protein significantly correlated with lymph node metastasis and depth of tumor invasion.This suggested that Gelsolin protein play an important role in gastric cancer invasion and metastasis.It can be used as biological indicators for assessment of invasion of gastric cancer metastasis.3. The expression of uPA in gastric cancer is higher than that in adjacent mucosa.The expression of uPA was correlated with the patient clinical pathological stage, tumor differentiation,tumor invasion depth and lymph node metastasis, suggesting that uPA can promote metastasis of gastric cancer,may be indicators for assessing tumor invasion and metastasis.4.
