ObjectiveTo detect the expression levels of BRMS1 protein and MMP-9 protein in breast cancer tissues by immunohistochemical method,and study the expression of BRMS1 and MMP-9 protein in normal breast tissues,benign breast disease,adjacent no cancerous breast tissues and breast carcinoma tissues to probe the clinical significance of them in different kind of breast tissues.And it will can provide Molecular Biology Detect indicators for promoting the level of diagnosis and treatment of breast cancer and improve prognosis evaluation through research of the two metastasis-associated gene expression and the relationship between different and laying the foundation of theory and practice for further studying BRMS1,this newly discovered metastasis suppressor gene.MethodsThe expression of BRMS1 and MMP-9 protein were detected by using EliVision immunohistochemical techniques in 80 cases of primary breast cancer,20 cases of adjacent non cancerous breast tissue,32 cases of benign breast disease and 10 cases of normal mammary gland tissues,and analysis the clinical significance with histopathologic records,and analysis the correlation between BRMS1 and MMP-9 in the same time.All data were processed by SPSS13.0, comparisons were done using chi-square test and Fisher's exact test,examination standard by two-sided testα=0.05. Results1.The immunostaining of BRMS1 localized mainly in nucleus and cytoplasm,showed diffuse distribution in cells.BRMS1 strongly expressed in all normal breast and benign breast disease tissues(the strongly positive or positive expression rate of both is same,100%,10/10, 32/32);the rate of positive expression in adjacent non cancerous breast tissues is 85.00%(17/20); The positive staining of quantity and intensity of BRMS1 protein in breast cancer tissues significantly reduced than in normal breast tissues,benign breast diseases and tumor adjacent breast tissues(P<0.01).The expression of BRMS1 protein is negative in most breast cancer cells, but show positive expression in some breast cancer tissues,and the rate of positive expression was 40.00%.2.The immunostaining of MMP-9 localized mainly in cytoplasm.It was not expressed in the normal breast tissues,and the rate of positive expression of MMP-9 protein in benign brease disease was 6.25%,and was 15.00%in adjacent noncancerous breast tissues,but it was 67.50%(54/80) in breast cancer tissues.The rate in breast cancer tissues significantly reduced than in normal breast tissues,benign breast diseases and tumor adjacent breast tissues(P<0.01)3.The positive expression rate of BRMS1 in breast carcinoma group>2cm in tumor diameter(33.33%,15/45) was lower than that in the group≤2cm in tumor diameter(48.57%, 17/35).There was no significant difference between them(P=0.168).The positive expression rate of BRMS1 in breast carcinoma of clinical stageⅢ(14.30%,4/28) was lower than in breast carcinoma of stageⅡ(54.55%,18/33) and stageⅠ(52.63%,10/19),and there was significant difference among them(P=0.003).The positive expression rate of BRMS1 in breast carcinoma group with lymph node metastasis(14.71%,5/34) was significant lower than that in breast carcinoma group without lymph node(58.70%,27/46),there was significant difference between them(P=0.000).The positive expression rate of BRMS1 in breast carcinoma group with positive expression of ER(52.63%,20/38) was significant higher than that in breast carcinoma group with negative expression of ER(28.57%.12/42),there was significant difference between them (P=0.028).The expression of BRMS1 in breast carcinoma did not correlate with patient's age, histological grading,histopathological classification and the station of PR.et al.(P=0.709; P=0.722;P=0.905;P=0.645). 4.The positive expression rate of MMP-9 protein in breast carcinoma group>2cm in tumor diameter(77.78%,35/45) was higher than that in the group≤2cm in tumor diameter (54.29%,19/35).There was significantly difference between them(P=0.026).The positive expression rate of MMP-9 protein in breast carcinoma of clinical stageⅢ(85.71%,24/28) was higher than in breast carcinoma of stageⅡ(69.70%,23/33) and stageⅠ(36.84%,7/19),and there was significant difference among them(P=0.003).The positive expression rate of MMP-9 protein in breast carcinoma group with lymph node metastasis(88.24%,30/34) was significantly higher than that in breast carcinoma group without lymph node(52.17%,24/46),there was significantly difference between them(P=0.001).The positive expression rate of MMP-9 protein in breast carcinoma of histological gradingⅡ(65.38%,17/26) andⅠ(50.00%,13/26) were higher than in histological gradingⅢ(85.71%,24/28).there was significantly difference between them(P=0.019).The expression of MMP-9 in breast carcinoma did not correlate with patient's age, histopathological classification and the station of ER and PR,et al.(P=0.845;P=0.238;P=0,430; P=0.508).5.The positive expression rate of MMP-9 protein was 75.00%(36/48) in the breast carcinoma group with negative expression of BRMS1,but it was 56.25%(18/32) when BRMS1 showed positive expression in the tissues.There was no significant correlation between the expressions of BRMS1 and MMP-9 in breast carcinoma(r=0.193,P=0.079)Conclusions1.The different expressions of BRMS 1 protein or MMP-9 protein in breast cancer and normal breast tissues and benign breast disease and adjacent non cancerous breast tissue had remarkable differences.Therefore the two proteins would be identified as tumor markers in order to diagnose breast cancer or other malignant tumors.2.The expression of BRMS 1 decreased in breast cancer;The decreased expression of BRMS1 did not correlate with patient's age,the size of breast tumor,histological grading,histopathological classification and the station of PR.et al.and significantly correlated with the stage of TNM of breast cancer,the lymph node metastasis and the station of ER protein expression.These indicate the decreased expression of BRMS1 may takes part in the regulation progression of breast cancer metastasis,and its contribution may correlate with ER.BRMS1 assessment can be used as indicators of the biological behavior of breast cancer3.The expression of MMP-9 increased in breast cancer;the increased expression of MMP-9 did not correlate with patient's age,histopathological classification and the station of PR and ER protein expression,et al.and significantly correlated with the size of breast tumor,histological grading,the stage of TNM of breast cancer and the lymph node metastasis.The rate of MMP-9 positive expression is higher with higher histological grading;progress with the clinical stage and higher;and the expression of MMP-9 protein with axillary lymph node metastasis were significantly higher than that without lymph node metastasis in breast cancer tissues,suggesting that MMP-9 expression may promote the metastasis of the process of breast cancer,the assessment can be used as indicators of the biological behavior of breast cancer.4.There was significant correlation between the expression of BRMS1 and MMP-9 in breast carcinoma,which indicated that expression imbalance of the two protein in breast cancer tissues may be a complementary manner to participate in the invasion process of metastasis of breast cancer.Joint detection in tumor tissues of both the expression of the two proteins may contribute to early diagnosis and prognosis.
