| Objective: Alzheimer's disease(AD) is a common and frequently occur -ring disease of older persons.For the treatment of senile dementia, there are no special effect methods and medicines at home and abroad. Chinese traditional medicine has a unique understanding of senile dementia of the etiology, pathogenesis. The clinical practice has accumulated a wealth of therapeutic experience in the long term. Chinese medicine attribution of dementia to stay in the chinese dementia, the text silly, forgetful, language reversed, and the crazy, depression syndrome and so on. We abide by the rule " tonifying kidney and invigorate spleen, clear phlegm stasis", Compound Rehmannia to nourish kidney Yin , Spleen Qi, clear phlegm and blood stasis, filling the role of the brain nerves, while compound recipe of Chinese medicine includes multi-composition, single-herbs also contains a variety of ingredients ,they can affect one or a variety of target sites, to study Compound Rehmannia effections of SAM-P/8 mice: to observe SAM-P/8 mice?s behavior changes; morphology tissue change by light microscopy, detecting Bcl-2, Bax protein expression in hippocampal neurons by using immunohistochemistry; apoptosis cells changes by flow cytometry machine.to observe furtherly Compound Rehmannia on Alzheimer's disease efficacy and mechanism of action, for providing theoretical guidance of the clinical using; for elaborating furtherly the pathogenesis of Alzheimer's pathology provide an experimental basis.Methods: SAM-P/8 mices were divided randomly into three groups:Com -pound Rehmannia group,western medicine group, model group, 15/group. Compound Rehmannia group was lavaged by Compound Rehmannia solution (1ml/day,individual). western medicine group was lavaged by Piracetam solution (1ml/only, 75tablets/300ml purified water), normal control group, the adoption of healthy Kunming mices, and the model group with normal saline 1ml/day gavage.After 3 weeks, to test each group mice behavior by Morris water maze;observe hippocampal neurons changes by light microscopy and electron microscopy;determin Bcl-2, Bax protein expre -ssion in hippocampal by immunohistochemistry;observe apoptotic cells by flow cytometry machine, then To make the datas for statistical analysis . Results:1.By Morris water maze test, Compound Rehmannia group SAM-P/8 mices escape latency 29.50±2.86 shortened , staying in the original platform 38.48±3.26 prolonged, which was significant difference (p<0.05) compared with model group 38.64±3.86,29.81±5.16, But no significant difference (p>0.05) compared with western medicine group.2.Morphological observation showed that the Compound Rehmannia group in the hippocampus and the frontal cortex neurons rules closely arranged, structured and number of glial cells was less than model group, ultrastructural changes was minor, the structure tends to the normal mice .3.The results of immunohistochemistry, the spectral area of Bcl-2 IOD value 16.64±2.27 and Bax value 0.16±0.014 of Compound Rehmannia group was significant difference (p<0.05) compared with the model group Bcl-2 IOD value 5.15±0.99; Bax IOD value 0.25±0.028.But no significant difference (p>0.05) compared with the western medicine group Bcl-2 IOD value 16.96±2.83; Bax IOD value 0.18±0.019.4.Apoptotic cells by flow cytometry analysis, normal group apoptosis rate is 3.19±0.12, model group apoptosis rate reached 16.28±2.22, while the Compound Rehmannia group apoptosis rate (10.96±1.84) was significant difference (p<0.05) compared with the model group and normal group.Conclusion: Compound Rehmannia can improve Alzheimer's mice learn -ing and memory ability, reduce brain tissue damage, and enhance brain neurons function, and Reduce the number of neuronal apoptosis by up-regulated anti-apoptotic gene Bcl-2 and down-regulated apoptotic gene Bax expression. It can prevent and care better the SAM-P / 8 mice brain neurons. |
