Diabetes,because of the body absolutely or relatively lack insulin and result of the sugar metabolic disordor. Its character as the high glucose level.Now,The western medicine in the treatment of diabetes is effective, but have damage to liver and kidney,also some side effects, and can not be effectively controlled the complications. The effect of the Chinese herbal is stable, mild and little secondry reaction, also can effectively prevent many complications.Radix Rehmannia is the dried or fresh block root of Rehmannia glutinosa Libosch,.Radix Rehmannia is one of beneficial traditional Chinese herbal,first records in the Shennong'herbal classic,now,recorded in the pharmacopoeia.Study on the chemical components and pharmacological activities demonstrated that iridoid glycosides and polysaccharide were the main chemical components and active principles of Rehmannia glutinosa Libosch.And the catalpol,is the highest content substance of the aniridoid glycoside.The catalpol has many activities,such as anti-cancer,,diuresis,laxness,hypoglyce mic activity and neuroprotection,was considered as one of the main effective compo nents.It has attracted much interest in researching on the extraction and isolation technology in recent years.Iridoid glucosides from Rehmanniae have large polarit y,similar structure and poor stability,so it is difficult to get a great amount of catalpol with the purity over 90%.Thus limited the bioactivity reserach and industrialize application.Our reserach was intended to investigate the enrichment and purification technology of catalpol,and evaluated the bioactivities of hypoglycemic activity. We used catalpol yield as the evaluation index. Based on the analytical assay of catalpol,we used orthogonal design to choose the best extraction technology of catalpol.We have compared ten kinds of macroreticular resins with different polarity for separation of catalpol,such as D101,HPD100,HPD600 and so on,and selectted the better one was Actived carbon.We used 95% ethanol to refluence the Actived carbon,and over 60% can be eluated down.We used the diabetic mice to evaluate the bioactivities of hypoglycemic activity.Th e result showed that catalpl could lower the glucose level in alloxan-induced diabetic mice and improve the glucose tolerance.We used the biochemistry kit to test the serum of diabetic mice.It showed that catalpol could improve the antioxidantion of the diabetic mice in a dose-dependent manner.
