The study aimed at a preliminary discussion of the genetic diversity of the white feather broiler breeds and the early work of molecular level breeding. Eight varieties materials were collected for testing.RAPD-PCR molecular marker technology was applied to explore the classification of the white feather broiler breeds and genetic diversity. The main conclusions were as follows:(1) The quality of template DNA was critical to the RAPD-PCR Test,through the experiment,high quality DNA was extracted.Amplification for RAPD-PCR Test was approved.(2) The technical system of RAPD-PCR reaction for white feather broiler breeds was established and optimized. A 20μl reaction system, with 1.5U rTaqDNA polymerase, 0.35μM primer,0.25mM dNTPs,40ng template DNA and 2μ1 10×PCR buffer was established. Under the system,5min pre-denaturation at 94℃,30s denaturation at 94℃,45s annealing at 37℃,90s extension at 72℃, for 35 cycles, 10min extension at 72℃, after the end of the cycle,4℃preserved. Amplified bands were clear, effective, and suitable for RAPD analysis.(3) In the test 28 primers,21 were selected for the white feather broiler breeds RAPD anplification. Including 114 polymorphism bands,162 bands were amplified. The polymorphism rate reached 70.37%.(4) According to the RAPD analysis, the genetic similarity values of eight varieties were between 0.484-0.740.(5) The white feather broiler breeds cluster map revealed genetic similarity between varieties, and more for the future purpose of seletive introduction, provided references for selected breeding and shortening the breeding period.
