AbstractThe classification position of 32 rices in intersubspecies of rice was initially established. Nine of the above rices were Hsien or Indica, thirteen were partial lndica, four were partial Keng or Japonica and six were Japonica. Agronomic character inspection on some upland rices was made. The regression analysis and line regrssion equation between the yield and 7 quantity characters were carried out.The preparation method of DNA from the silica-gel-dried leaves of rice and upland rice was studied. The DNA with the length of 45 kb and yield of 50?170 ~t g/g have been acquired, which could be used successfully for conducting PCR. RAPD PCR was run in a reaction volume of 20 t.t L containing 50---- 100 ng template DNA, 1.2 Unit of Taq DNA polymerase, 2.5 mmol/L Mgcl2, 250 ii mol/L dNTPs, 0.5 ii mol/L primer, 10 mmol/L Tris-HC1(pH 8.0), 50 mmol/L KCI and 0.1% gelatin.42 DNA samples were amplified for random amplified polymorphic DNA analysis with 10 random primers, and 93 polymorphic bands were obtained. The amplified fragments ranged from 0.2 to 3 kb and the genetic distance of them ranges from 0.11 to 0.75. The fingerprints of 42 rices was getted. The result of the finger profile analysis and UPGMA cluster analysis indicated that RAPD only can identify rice species to some extent, but can't be intersubspecific marker. 42 rices can be differentiated from each other with 560, S80, Si 308 and S 1319. The acquisition of special bands of rice and upland rice, intersubspecific groups and all types of ecological species was failed. Bands of about 1.0 kb, 800 bp and 2.7 kb conducted by S80 and S 1308 were special marker bands of ew-varieties? Bands of some 1.5 kb and 2.5 kb respectively by S80 and Si 319 was special marker bands of lndica and Xiangzhenai? respectively.
