Strawberry, as one of the seven wonders of the world fruit, with high nutritive value, beautiful appearance and delicious taste, is known as the "Queen of Fruits". After30years of development, our country strawberry cultivation area and production have been the largest in the world. Most of the cultivars used in the production are from Europe, America and Japan, due to the different ecological conditions and consumer habits, introduced species are often not suited to the cultivation conditions in our country, neither fully meet our people’s spending habits. Tests showed that the introduction of Japanese varieties of quality but not disease-resistant, Europe and America varieties of high disease-resistant but not quality, so species selection fall into dilemma. Therefore on the basis of our country local strawberry germplasm begin to innovate, select disease-resistant, high sugar concentrated flavor and quality strawberry varieties will have the great significance to the development of our strawberries.Because strawberry is frequently introduced between different countries and regions makes varieties of confusion, which leads to some synonym and homonym phenomenon. Then causes the difficulties of strawberry collection and utilization and hindered the strawberry germplasm resource and varieties breeding. This experiment uses ISSR technology to identify specific varieties of strawberries then marks the establishment of the genetic tree. In addition, by34strawberry varieties genetic diversity analyses, reveals the genetic relationship between the varieties, offeres the technical support of strawberry varieties identification and crossing parents selection. The results are as follows:(1) The DNA of strawberry leaves using a modified CTAB method can be used for ISSR markers.(2) The suitable strawberry ISSR analysis system and amplification reaction program had established.(3)11out of96ISSR primers can be used for34strawberry resources of the DNA polymorphism detection, Constructs the genetic clustering tree diagram.(4) During similarity coefficient of0.75,34strawberries classified into seven groups then analyze the genetic relationship between the varieties.(5) Genetic similarity coefficient between Japan varieties(Toyonoka、Benihoppe、 Akihime)and Europe and America varieties(Sweet charlie、Tudla、Camarosa) is0.71to0.82, the genetic relationship between them in medium remote, appropriate configuration combinations.To breed with high soluble solid content, strong ability of resistance to Powdery Mildew in fine varieties for breeding objective, from the hybrid with high quality Japanese varieties and high yield Europe and the United States varieties select9hybrid combinations. Finally, from hybrid progeny plants, we selected high-quality, disease-resistant, high-yielding germplasm two, high quality germplasm three, disease-resistant germplasm three.
