Iris halophila Pall.is a kind of perennial herbaceous plant of Iris L. It has a lot of characteristics such as fighting a cold, a drought, and insect pests, strongly adaptability and so on. So it Is the resistance of genetic resources and breeding of fine landscape, excellent alkali soil improvement plants. At present, Iris halophila Pall.is still in a state of undeveloped. With the influences of environment changing and human activity, their genetic diversities have risk of comedown. In this article, the genetic diversity and genetic relationship of Iris halophila Pall. populations in Xinjiang were studied by SSR and SRAP markers.(1)6 pairs screened from 10 primers of SSR and 18 pairs of SRAP selected out from 120 primers were used to analysis the genetic diversity of Iris halophila Pall. in Xinjiang.45 bands were amplified from 9 natural populations by 6 reliable primers.The percentage of polymorphic were 100%.The Nei's gene diversity index was 0.2949 and the Shannon diversity index was 0.4672.The gene diversity within population was 0.2468,71.58% of genetic variation existed within populations,while 28.42% of genetic variation existed among populations.The genetic differentiation coefficient was 0.2842.The gene flow was 1.2593.112 bands were amplified from 9 natural populations by 18 reliable primers.The percentage of polymorphic were 100%.The Nei's gene diversity index was 0.3628 and the Shannon diversity index was 0.5433.The gene diversity within population was 0.3194,75.31% of genetic variation existed within populations,while 24.69% of genetic variation existed among populations.The genetic differentiation coefficient was 0.2469.The gene flow was 1.5251.(2) Analysis of molecular variance (AMOVA) further revealed that 31.14% of SSR or 28.36% of SRAP variation resided among population.(3) Clonal diversity was also high with Simpson deversity index (D) were 0.9901 of SSR and 0.9872 of SRAP at species level.This may indicate that multi-clone origin of populations, and clonal reproduction also has an important influence on the genetic structure of Iris halophila Pall..(4) Calculated the genetic simicarity coefficient between 270 accessions base on Nei & Li's method with the datas of SSR, SRAP and their aggregation.The result of clustering base on UPGMA indicated that populations with similar ecological and geographical conditions clustered first.(5) The correlation between the two sets of similarity parameters calculated with SSR and SRAP data respectively (r=0.9971,p< 0.001, indicating that the two methods were consistant and able for detecting genetic diversity of Iris halophila natural populations in Xinjiang.
