V.davidii.Foex is a wild grape species originated from China,it has the characteristics of resistance to high temperaturea and high humidity,strong resistance to fungal diseases.Meanwhile,the grape fruit has a appropriate content of sugar and acid,rich nutrition and a huge potential of fresh and processing.The genetic diversity study of grape germplasm is benefit to the collection,conservation,identification,germplasm innovation and effection utilization of V.davidii.Foex germplasm.This research combines botany property descriptors and SRAP markers,study the genetic diversity of 92 V.davidii.Foex germplasm materials by hierarchical cluster analysis,principal components analysis,aimed to identify the genetic relationship among wild V.davidii.Foex germplasm in China.The main results were as follows:1.Through the characterization of V.davidii.Foex germplasm botany traits,the basic statistical results show that:27 character variables have the average variation coefficient of 30.06%.Among the 27 character variables,prostrate hairs color on node of the tip has the biggest variation coefficient of 81.33%;luster of the upper side of young leaf,prostrate hair variety of the shoot,prostrate hair dense of the shoot and shape of upper sinuses of mature leaf have variation coefficient of 0.2.The clustering analysis of botany traits show:92 grape germplasms have the similarity coefficient range of 0.22-1.00.The grape germplasms are basically clustered into two big groups,the first category includes all Jiangxi grapes and Se grape(V.davidii.Foex),the second includes the rest Hunan grapes.The results of principal component analysis show the first principal component contribution rate is 13.38%,and accumulative variance contribution ratio of the first three principal components is 32.44%.3.A total of 100 pairs of primer combination are used in this study,22 screened primers,which have obvious difference stripes and can repeat the differences,produced 182 clear bands,of which 157 bands are polymorphism bands;Each primer combination of grape material has amplified polymorphism ratio between 78.0%-91.0%,average polymorphism ratio is 86.3%.The 92 grape germplasms have similarity coefficient between 0.32-0.98.Amplified fragment size is concentrated between 100-1000bp.SRAP markers cluster analysis,92 germplasm materials are cluster into 2 big groups;The results of principal component analysis show cumulative variance contribution ratio of the first three principal components is 39.44%,the first principal component varianc contribution rate is 21.93%.4.Using NYSTSpc2.10 software to carry Mantel test on the cluster analysis results of botany descriptors and SRAP markers,it has the correlation coefficient r is 0.5298,shows that the classification of those two markers has high consistency.Based on the analysis of two markers,92 grape germplasms have abundant genetic diversity,different regions have far genetic distance,seven V.davidii.Foex of Hunan have close genetic distance,some of Jiangxi province V.davidii.Foex almost overlapping.Based on the analysis,this study establish a botany properties and SRAP marker NYSTS database,which can be used to identify new discovery wild V.davidii.Foex germplasm,and analysis its genetic relationship with the existing germplasm.
