In this paper, the chemical compositons and their structures in the leaves of Persimmon were described. The extracting methods, isolation and identification of Persimmon were also introduced. First, we investigated the optimum extracting condition of flavonoids from the leaves by using the orthogonal design. And the results showed 60 °C ,40:1 of 45% ethand to the leaves weight ratio, extracting 5h were optimal. In this condition, we extracted the flavonoids and measured its content.the result indicated the average content is 1.5% . Second, we used three ways to purify the Persimmon flavonoids. The three ways separately were aqueous two-phase extrction technology, supercritical fluid technology, and ceramic membrane filtration. After comparison, we finded the best ways for purification of Persimmon flavonoids was aqueous two-phase extrction technology. Flavonoids of persimmon leaves were recovered with high yield and high concentration in the top polymer-rich phase at 25%PEG 600 and 25%(NH4)2SO4 were applied . The suitable extraction conditions were pH 11, MgCl2 3%,temperature 25 °C. Third, we can obtain solid Persimmon flavonoids through acid deposition and dryness. Fourth, the solid Persimmon flavonoids was detected by infraed spectroscopy technology, high performance chromatography(HPLC) and matrix-assisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF MS). The result showed the solid Persimmon flavonoids contained rutin and other flavonoids.
