Supercritical Fluid Extraction Method is a newly developed technology forextracting and separating substance.Among them, the supercritical CO2is akind of supercritical fluid which the most commonly used.Extracted the naturalproduction with supercritical CO2have the effective abilities of extraction andseparation ,and have many advantages of gathering extract, separate andcondense for the integral,and the whole operation cost is low, without solventremaining, freeing from pollution, economizing the energy, the operationconvenience etc.which have already been applied in many realms at homeand abroad, such as food, spice, petroleum, chemical engineering and themedicine industry etc.The method which replace the traditional solvent withsupercritical CO2 has been widespread acceptde by the industryfield.American environment protection bureau( EPA) already gradually regardthe supercritical fluid extraction as the standard method to replace thetraditional solvent, especially for the Chinese herbal medicine of the volatileoil or the liposolube substance, However ,nowadays still didn't have goodmethod and the production equipments of industrialization. Which has alreadyserious restricted the development and the production of this kind ofmedicine.However the supercritical CO2 extraction have the obviousadvantage in this aspect comparing with traditional Chinese herbal medicine,and have the good industrial prospect.Many medical experts put forward andadopt the advantange technology of supercritical extract many times to carryon the Chinese herbal medical research development and industrilization.Beshowed from this, the supercritical fluid extraction is a kind of trend for thetechnical development, for increasing the medical level of Chinese herbalmedicine system , particularly is to solve the difficulties of extract andseparate volatility and the lipiosolube of medicine composition efficently,having the very strong practicability, urgency and necessities.Therefore,adopting the supercritical CO2 technique to extract is the reform to thetraditional Chinese herbal medicine,and have huge applied value to theChinese herbal medical industry high technique development.In this article the author mainly discusses about the application of thismethod in extracting effective contents from herbs In this article the authortakes the extraction of Cnidium monnieri ,Psoralea Corylifolia, Angelicadahurica and Ligusticum chuanxiong as the example.Traditional extrationmethod of the valid composition of Chinese herb medicine is cooked bywater,extracted by ethanol or ligroin, but the traditional methods havesome defect,such as insecurity in operation ,the low ablitily of extraction, thevalid composition lose severely, the quantity is unsteady,and hard assure thefunction of valid composition effecently , and the more important is that resultin medical pollution and pollution of the environment easily.However theadoption of supercritical extraction have the advantange of the credibileoperation and effect result of separating that can overcome the traditionalweakness, and Extraction Method can guarantee safer operations andbetter extraction output rate. Supercritical Fluid Extraction Method(abbreviated as SFE) can be used to extract effective contents from naturalherbs, which takes shorter time, uses less materials and lower costs, undermore controllable conditions. It would not cause any pollution or destruction ofthe sample products.For that purpose, the author first discuss the technological conditions forextracting by Supercritical Fluid Extraction Method, the author takes theextraction of Cnidium monnieri , Psoralea Corylifolia, Angelica dahuricaand Ligusticum chuanxiong as the example. and worked out the relationshipbetween operation pressures and extraction output rates. It has been provedthrough tests that the density of Supercritical Fluid becomes higher with thepressure goes higher and then its dissolving rate rise respectively, and sodoes the extraction rate. But once it reaches a certain pressure, the increaseof extraction rate becomes less and less obvious. So a conclusion can bedrawn that the ideal pressure range is 20-40Mpa. The temperature of fluidalso effects the extraction rates, that is, the dissolving rate of extractionsolvent goes higher with the temperature, and then the extraction ratebecomes larger, but after reaching a certain temperature the extraction rategoes down. Experiments show that the suitable temperature range forextraction operation is 35--70℃. It is also manifested that the best operationtime of extraction should be about 2~6 hours, or so.Meanwhile tests havebeen done on the effects of addictive to extraction rate. HPLC was applied to determine the contents of the totalcoumarins.Range analysis and variance analysis were applied to optimizeSFE process,which put forward the scientific and reliable method for extractthe valid contents from coumarin material substance of Chinese herbmecicine by supercritical CO2 fluid.The thin -layer chromatography wasapplied to identify the SFE extractives qualitive.;And the ultravioletspectrophotometric method was applied to meature the total validcomposition extracted by SFE;Built up the method of high performanceliquid chromatography to measure the the total valid composition ofextractives by SFE ,which supply the feasible methods for quantitymeasures.And adopt the most simple method -ethanol crystallize method toseparate and obtain Cnidium monnieri crystal and Psoralea Corylifoliacrystal.And adapt the four greatest spectrums ( UV,IR,HNMR,MS) to 1obtain the chemistry data to reflect chemistry composition information of thetwo crystals, further made sure its chemistry structure. At the same time in this article the author combine the advancesupercritical CO2 extraction technique with separated and refined processorganically, such as the extraction of Cnidium monnieri and PsoraleaCorylifolia , is to adopt supercritical extraction technique and crystallizetechnique etc. gathers excellent;And such as the extraction of Angelicadahurica , adopting the supercritical extraction technique and technique usebig bore resins to adsorb and separate substances.Therefore thesetechniques make reserved rate of the valid composition raised with maximumlimit and the category of valid composition is complete,which make theprocess of separate to attain the real optimition. And in this article the authors make use of the supercritical CO2extraction technique to extract and research the medicinal materials ,forexample Cnidium monnieri , Psoralea Corylifolia, Angelica dahurica and...
