Flos.lonicerae is a Chinese traditional medicine, which have several benefits to humans, including antipyretic ,antibacterial and detoxifcation function. Its main active ingredients are chlorogenic acid, isochlorogenic acid and flavone etc. In the article, the extraction progress of chlorogenic acid, HPLC fingerprint analysis and the bacteriostatic test of extract were studied.In the study on Flos, lonicerae extraction technology, according to extraction yield and the yield of chlrogenic acid, solvent extraction. ethanol precipitation. ethanol extraction and enzymatic treatment were investigated in detail. The results showed that using chlrogenic acid as criterion, enzymatic treatment was the best, the yield can be up to 12.82%, then the next was ethanol extraction; and using extraction yield as standard, enzymatic treatment was the best as well, up to 37.63%, while the others were worse. So the effect of enzyme dosage, treatment time and treatment temperature on extraction yield were discussed. Under optimal condition, extraction yield was 37.63% and the yield of chlrogenic acid was 12.82%. Enzymatic treatment time and dosage had the same effect, that was to say-the effect of increasing treatment time was identical to increasing enzyme dosage , and the optimal temperature of enzymatic treatment was 40癈 to 50癈.In the study on HPLC fingerprint analysis of extractions, the mobile phrase such as water -acetonitrile -acetic, methanol-water- acetic, methanol - sodium acid phosphate(10%), and methanol-sodium acid phosphate(0.1mol/l) were tested respectively , the results showed that methanol-water - acetic(30:70:0.5) was optimal , the peak of chlrogenic acid was separated from others . Under this condition to detect chloregnic acid at 297nm. 310nm. 325nm, the wavelength 325nm was optimal. Reserved time of chloregnic acid was 4.2 min. According to sequence all the chromatogram peaks were ranked and the relevant area Ar were calculatedAr=area of the peak of sample /total area of criterion On basis of the preview formula, HPLC fingerprint of Flos loniceraewere established to distinguish the category, different extraction methods and distinguish Flos Lonicerae from other species.In the bacteriostatic test, thirteen different bacterium was experimented. Results showed that the extract was more effective against staphylococcus aures and coliform. The MIC of extract from Flos Lonicerae against staphylococcus aures was 6.25mg/ml, based on chlrogenic acid the MIC was 1.81 mg/ml , and the MIC against coliform was 12.50mg/ml, based on chlrogenic acid the MIC was 3.62 mg/ml...
